One of the important determinants of virulence of Mycobacterium
tuberculosis is adaptation to adverse conditions encountered in the host
cells. The ability of Mycobacteriumto successfully adapt to stress condi-tions is brought about by the expression of specific regulons effected by a
repertoire of rfactors.
A series of trehalose-based oligosaccharides were isolated from the cytoplasmic fraction of Mycobacterium smegmatis and puriﬁed by gel-ﬁltration and paper chromatography and TLC. Their structures were determined by HPLC and GLC to determine sugar composition and ratios, MALDI-TOF MS to measure molecular mass, methylation analysis to determine linkages, 1H-NMR to obtain anomeric conﬁgurations of glycosidic linkages, and exoglycosidase digestions followed by TLC to determine sequences and anomeric conﬁgurations of the monosaccharides....
The Mycobacterium tuberculosisgenome contains an
unusually high number of proteins involved in the metabo-lism of lipids belonging to the Lip family, including various
nonlipolytic and lipolytic hydrolases. Driven by a structural
genomic approach, we have biochemically characterized the
Rv1399c gene product, LipH, previously annotated as a
putative lipase. Rv1399c was overexpressed inE. colias
inclusion bodies and refolded. Rv1399c efficiently hydro-lyzes soluble triacylglycerols and vinyl esters.
The trehalose-phosphate synthase (TPS) ofMycobacterium
smegmatiswas previously purified to apparent homogeneity
and several peptides from the 58 kDa protein were
sequenced. Basedon that sequence information, the gene for
TPSwasidentified inthe Mycobacterium tuberculosis
genome, and the gene was cloned and expressed inEscheri-chia coliwith a (His)6tag at the amino terminus.

So, in summary, traction is important for transmitting the forces from the wheels to the
