DNA replication, the process of copying one double stranded DNA molecule to produce two identical copies, is at the heart of cell proliferation. This book highlights new insights into the replication process in eukaryotes, from the assembly of pre-replication complex and features of DNA replication origins, through polymerization mechanisms, to propagation of epigenetic states. It also covers cell cycle control of replication initiation and includes the latest on mechanisms of replication in prokaryotes. The association between genome replication and transcription is also addressed.
The faithful and complete replication of
chromosomes is essential to maintain
genetic stability and prevent the accumulation
of cancer-promoting mutations.
Replication forks are vulnerable to stalling
or collapse as they encounter obstacles on
the DNA template, which can be unrepaired
DNA damage, DNA-bound proteins or
secondary structures. Similarly, chemical
agents like hydroxyurea and aphidicolin
inhibit replication elongation, leading to
fork stalling or collapse.
Meiotic recombination is initiated by DNA double-stranded breaks intro-duced by the SPO11 protein. Despite a decade of research, the biochemical
functions of SPO11 remain largely unknown, perhaps because of difficulties
in studying the functionally active SPO11.
The occurrence of DNA double-strand breaks in the nucleus provokes in
its structural organization a large-scale alteration whose molecular basis is
still mostly unclear. Here, we show that double-strand breaks trigger pref-erential assembly of nucleoproteins in human cellular fractions and that

For the first time, the detection of DNA sequences, related to HIV (found in AIDS patients), using Methylene Blue (MB) as the redox indicator was performed. The voltametric signals have been investigated on bared chitosan (CS) modified; probe immobilized (DNA single strand) and target-hybridized (DNA double strand) electrodes sequentially by means of Cyclic Voltammetry (CV) and Square Wave Voltammetry (SWV).
