DNA replication, the process of copying one double stranded DNA molecule to produce two identical copies, is at the heart of cell proliferation. This book highlights new insights into the replication process in eukaryotes, from the assembly of pre-replication complex and features of DNA replication origins, through polymerization mechanisms, to propagation of epigenetic states. It also covers cell cycle control of replication initiation and includes the latest on mechanisms of replication in prokaryotes. The association between genome replication and transcription is also addressed.
The faithful and complete replication of
chromosomes is essential to maintain
genetic stability and prevent the accumulation
of cancer-promoting mutations.
Replication forks are vulnerable to stalling
or collapse as they encounter obstacles on
the DNA template, which can be unrepaired
DNA damage, DNA-bound proteins or
secondary structures. Similarly, chemical
agents like hydroxyurea and aphidicolin
inhibit replication elongation, leading to
fork stalling or collapse.
Meiotic recombination is initiated by DNA double-stranded breaks intro-duced by the SPO11 protein. Despite a decade of research, the biochemical
functions of SPO11 remain largely unknown, perhaps because of difficulties
in studying the functionally active SPO11.
The occurrence of DNA double-strand breaks in the nucleus provokes in
its structural organization a large-scale alteration whose molecular basis is
still mostly unclear. Here, we show that double-strand breaks trigger pref-erential assembly of nucleoproteins in human cellular fractions and that
they mediate the separation of large protein–DNA aggregates from aque-ous solution.
For the first time, the detection of DNA sequences, related to HIV (found in AIDS patients), using Methylene Blue (MB) as the redox indicator was performed. The voltametric signals have been investigated on bared chitosan (CS) modified; probe immobilized (DNA single strand) and target-hybridized (DNA double strand) electrodes sequentially by means of Cyclic Voltammetry (CV) and Square Wave Voltammetry (SWV). The response of hybridization between the probe and perfectly complementary, double-base mismatch, single-base mismatch of analyzed targets were clearly identified.
Chapter 6 - DNA structure, replication, and recombination. This chapter includes contents: Experimental evidence for DNA as the genetic material, the Watson and Crick double helix model of DNA, genetic information in DNA base sequence, DNA replication, recombination at the DNA level.
DNA repair must take place within the context of chromatin, and it is
therefore not surprising that many aspects of both chromatin components
and proteins that modify chromatin have been implicated in this process.
One of the best-characterized chromatin modification events in DNA-dam-age responses is the phosphorylation of the SQ motif found in histone
H2A or the H2AX histone variant in higher eukaryotes. This modification
is an early response to the induction of DNA damage, and occurs in a wide
range of eukaryotic organisms, suggesting an important conserved func-tion....
Concept of hybrid molecules:
When double strand DNA is steamed to a temperature
exceed the melting temperature (Tm), it will separate
into 2 single strands DNA due to breaks of H bonds. If
the reaction temperature is then decreased slowly plus
other appropriate experimental conditions, these
ssDNA will pair again. This phenomenon is called the
hybridization of molecules.
Characteristic of the hybrid molecules: Specificity, the
re-pairing occurs only between two complementary
The length of chordae was a measurement
of the distance between the head of the PM and the free edge of the posterior leaflet. Length
of the opposing chordae of the posterior leaflet was measured directly by using a calliper.
Double-armed mattress sutures with CV-5 GORE-TEX were placed at the fibrous tip of the
PM using PTFE on both sides and tied down firmly. In all cases Doi performed Duran ring
annuloplasty. Thereafter, the ePTFE suture is placed through the anterior leaflet.
Homologous recombination (HR) serves to eliminate deleterious
lesions, such as double-stranded breaks and interstrand crosslinks,
from chromosomes. HR is also critical for the preservation of replication
forks, for telomere maintenance, and chromosome segregation
in meiosis I. As such, HR is indispensable for the maintenance
of genome integrity and the avoidance of cancers in humans. The
HR reaction is mediated by a conserved class of enzymes termed

The beginning of the new millennium was marked by the announcement that the vast majority of the human genome had been sequenced. This milestone in the exploration of the human genome was preceded by numerous conceptual and technological advances. They include, among others, the elucidation of the DNA double-helix structure, the discovery of restriction enzymes and the polymerase chain reaction (PCR), the development and automatization of DNA sequencing, and the generation of genetic and physical maps by the Human Genome Project (HGP).
Second, board members in these small organizations often do double duty. One mo-
ment they are directors who set policy and supervise the executive. The next moment,
they are volunteer staff who greet new clients, stuff envelopes, or run the phone tree for
the annual fundraiser. This is an inherent and unavoidable conflict of interest (though
it is manageable). The board members are supervising the executive in their policy role,
and doing the executive’s bidding in their staff volunteer role.
