  • Sugar conjugation is a major pathway for the inactivation and excretion of both endogenous and exogenous compounds. We report here the molecular cloning and functional characterization of a phenol UDP-glucosyltransferase (UGT) from the silkworm, Bombyx mori, which was named BmUGT1. The complete cDNA clone is 1.6 kb, and the gene is expressed in several tissues of fifth-instar larvae, including fat body, midgut, integument, testis, silk gland and haemocytes. The predicted protein comprises 520 amino acids and has  30% overall amino-acid identity with other members of the UGT family.

  • Uridine-5¢-diphospho-1-a-D-glucose (UDP-Glc) is a common substrate used by glucosyltransferases, including certain bacterial toxins such as Toxins A and B from Clostridium difficile. Fluorescent analogs of UDP-Glc have been prepared for use in our studies of the clostridial toxins. These compounds are related to the methylanthraniloylATP compounds commonly used to probe the chemistry of ATP-dependent enzymes. The reaction of excess methylisatoic anhydride with UDP-Glc in aqueous solution yields primarily the 2¢ and 3¢ isomers of methylanthraniloyl-UDPGlc (MUG).

  • The kinetic data on sugarcane (Saccharumspp. hybrids) sucrose synthase (SuSy, UDP-glucose: D-fructose 2-a-D-glucosyltransferase, EC are limited. We character-ized kinetically a SuSy activity partially purified from sugarcane variety N19 leaf roll tissue. Primary plot analysis and product inhibition studies showed that a compulsory order ternary complex mechanism is followed, with UDP binding first and UDP-glucose dissociating last from the enzyme.

  • Glycogen phosphorylases (GPs) constitute a family of widely spread catabolica1,4-glucosyltransferases that are active as dimers of two identical, pyridoxal 5¢-phosphate-containing subunits. In GP fromCorynebacterium callunae, physiological concentrations of phosphate are required to inhibit dissociation of protomers and cause a 100-fold increase in kinetic stability of the functional quarternary structure.
