The pumping action is accomplished using a positive pressure to push the liquid from the pump tank into the return line. The Liqui-Mover pump can handle high temperature and high pressure condensate without difficulty. Liqui-Mover pumps are available in nine sizes.   • Sulfur is the element with the largest number of solid allotropes. Most of these consist of unbranched cyclic molecules with ring sizes ranging from 6 to 20. In addition, polymeric allotropes are known which are believed to consist of chains in a random coil or helical conformation. Furthermore, several high-pressure allotropes have been characterized. In this chapter the preparation, crystal structures, physical properties and analysis of these allotropes are discussed.

  • We show a simple and reliable method of tRNA aminoacylation with natural, as well as non-natural, amino acids at high pressure. Such specific and noncognate tRNAs can be used as valuable substrates for protein engineering. Aminoacylation yield at high pressure depends on the chem-ical nature of the amino acid used and it is up to 10%.

  • The high-pressure stopped-flow technique is applied to study the CO binding in cytochrome P450cam (P450cam) bound with homologous substrates (1R-camphor, camphane, norcamphor and norbornane) and in the substrate-free protein. The activation volume DV # of the CO on-rate is positive for P450cam bound with substrates that do not contain methyl groups. The kon rate constant for these substrate complexes is in the order of 3 · 106 M)1Æs)1.

When com-pared with apo-enolase, and Mg-enolase, the Mn-enzyme differs from the others in three ways. Exposure to hydro-static pressure does not inactivate the enzyme.   • Populations in Southeast AsiaandSouthChinahave highfrequencies ofa-thalassemia caused by a-globin gene mutations and/or deletions. This study was designed to find an efficient and simple diagnostic test for the mutations and deletions. A duplex polymerase chain reaction (PCR)/denaturing high-pressure liquid chromatography (DHPLC) was used to detect the mutations and deletions. A blinded study of 110 samples, which included 92 a-thalassemia samples with various genotypes and 18 normal DNA samples, was carried out by the methods.

However, all of the measurements in their study were made after the release of pressure.   • Small-angle neutron scattering experiments were performed on horse azidometmyoglobin (MbN3) at pressures up to 300 MPa.Other spectroscopic techniques have shown that a reorganization of the secondary structure and of the active site occur in this pressure range. The present measurements, performedusingvarious concentrations ofMbN3, showthat the compactness of the protein is not altered as the value of its radius of gyration remains constant up to 300 MPa.
