The techniques required to allow cells to grow and be maintained outside the body have been developed throughout the 20th century. In the 50 years since the publication of the first human cancer cell line, HeLa (1), thousands of cell lines representing most of the spectrum of human cancer have been derived. These have provided tools to study in depth the biochemistry and molecular biology associated with individual cancer types and have helped enormously in our understanding of normal as well as cancer cell physiology.

  • Heavy Chain Diseases The heavy chain diseases are rare lymphoplasmacytic malignancies. Their clinical manifestations vary with the heavy chain isotype. Patients secrete a defective heavy chain that usually has an intact Fc fragment and a deletion in the Fd region. Gamma, alpha, and mu heavy chain diseases have been described, but no reports of delta or epsilon heavy chain diseases have appeared. Molecular biologic analysis of these tumors has revealed structural genetic defects that may account for the aberrant chain secreted.

  • THE TERM CELL IS DERIVED from the Latin cella, meaning storeroom or chamber. It was first used in biology in 1665 by the English botanist Robert Hooke to describe the individual units of the honeycomb-like structure he observed in cork under a compound microscope. The “cells” Hooke observed were actually the empty lumens of dead cells surrounded by cell walls, but the term is an apt one because cells are the basic building blocks that define plant structure.

  • To establish stable cell lines that produce recombinant multisubunit proteins, it is usually necessary to cotransfect cells with several independent gene constructs. Here, we show that a stepwise fusion of individually transfected cells, results in a fused cell-line that secretes a complete multisubunit protein. Functional expression of recombinant multisubunit proteins may require a defined expression ratio between each protein subunit. The cell-fusion technology described allows a predefined expression level of each subunit.
