  • Extracellular signal-regulated kinases (ERKs) have been shown to be acti-vated by opioids and functionally linked to addiction. Morphine-associated changes in ERK activity seem to be the characteristic features of opioid action. In this study, we observed a rapid and severe increase in ERK1⁄2 activity after a 5 min morphine treatment of HEK-MOR cells (transfected with the ratl-opioid receptor MOR1) expressing l-opioid receptor.

  • Opioid analgesics such as morphine and meperidine have been used to control moderate to severe pain for many years. However, these opioids have many side effects, including the development of tolerance and dependence after long-term use, which has limited their clinical use. We previously reported that mutations in the muopioid receptors (MOR) S196L and S196A rendered them responsive to the opioid antagonist naloxone without altering the agonist phenotype. In MORS196A knock-in mice, naloxone and naltrexone were antinociceptive but did not cause tolerance or physical dependence.

  • The gene encoding the mu-opioid receptor (OPRM1) has been reported to associate with a range of substance dependence and HRH2 is the gene coding histamine H2 receptor, the site of action for various compounds, e.g. amitriptiline and mianserin, used in the treatment of psychiatric disorders.
