The present study aimed to investigate the role played by the leaflets of the
plasma membrane in the uptake of drugs into cells and in their extrusion
by P-glycoprotein and multidrug resistance-associated protein 1. Drug
accumulation was monitored by fluorescence resonance energy transfer
from trimethylammonium-diphenyl-hexatriene (TMA-DPH) located at the
outer leaflet to a rhodamine analog.

K-Ras is a small G-protein, localized mainly at the inner leaflet of the
plasma membrane. The membrane targeting signal of this protein consists
of a polybasic C-terminal sequence of six contiguous lysines and a farnesyl-ated cysteine. Results from biophysical studies in model systems suggest
that hydrophobic and electrostatic interactions are responsible for the
membrane binding properties of K-Ras.
Caveolae are nearly ubiquitous plasma membrane domains that in adipo-cytes vary in size between 25 and 150 nm. They constitute sites of entry
into the cell as well as platforms for cell signalling. We have previously
reported that plasma membrane-associated caveolae that lack cell surface
access can be identified by electron microscopy.
Gangliosides are sialic acid-containing glycosphingolipids present on mam-malian plasma membranes, where they participate in cell-surface events
such as modulation of growth factor receptors and cell-to-cell and cell-to-matrix interactions. Antibodies to gangliosides have been associated with
a wide range of clinically identifiable acute and chronic neuropathy syn-dromes.
In the yeast Saccharomyces cerevisiae, plasma membrane H
activated by d-glucose. We found that in the absence of glucose, this
enzyme forms a complex with acetylated tubulin. Acetylated tubulin usu-ally displays hydrophilic properties, but behaves as a hydrophobic com-pound when complexed with H
-ATPase, and therefore partitions into
a detergent phase.
As an adaptation process to the growth retardation provoked by the presence of nonlethal concentrations of ergosterol biosynthesis inhibitors, Ustilago maydis alters the ratio of linoleic to oleic acid bound to plasma membrane ´ complex lipids [Hernandez, A., Cooke, D.T., Lewis, M. & Clarkson, D.T. (1997) Microbiology 143, 3165–3174]. This alteration increases plasma membrane H+-ATPase hydrolytic activity.

plasma membrane and cellular storage vesicles increase the level of toxic
pro-oxidative cytosolic dopamine.
The drug concentration inside multidrug-resistant cells is the outcome of
competition between the active export of drugs by drug efflux pumps, such
as P-glycoprotein (Pgp), and the passive permeation of drugs across the
plasma membrane. Thus, reversal of multidrug resistance (MDR) can occur
either by inhibition of the efflux pumps or by acceleration of the drug per-meation.

We havemade a comprehensive and quantitative analysis of
the lipid composition of caveolae from primary rat fat cells
and compared the composition of plasma membrane inside
and outside caveolae. We isolated caveolae from purified
to disrupt the membrane, or extraction with nonionic
detergent, followed by density gradient ultracentrifugation.
The carbonate-isolated caveolae fraction was further
immunopurified using caveolin antibodies.
