Motor neuron degeneration in amyotrophic lateral sclerosis involves oxida-tive damage. Glutathione (GSH) is critical as an antioxidant and a redox
modulator. We used a motor neuronal cell line (NSC-34) to investigate
whether wild-type and familial amyotrophic lateral sclerosis-linked G93A
mutant Cu,Zn superoxide dismutase (wt⁄G93ASOD1) modified the GSH
pool and glutamate cysteine ligase (GCL), the rate-limiting enzyme for
Glutathione (GSH) is an intracellular antioxidant synthesized from gluta-mate, cysteine and glycine. The human erythrocyte (red blood cell, RBC)
requires a continuous supply of glutamate to prevent the limitation of GSH
synthesis in the presence of sufficient cysteine, but the RBC membrane is
almost impermeable to glutamate.

Nitrogen monoxide (NO) is a cytotoxic eﬀector molecule produced by macrophages that results in Fe mobilization from tumour target cells which inhibits DNA synthesis and mitochondrial respiration. It is well known that NO has a high aﬃnity for Fe, and we showed that NO-mediated Fe mobilization is markedly potentiated by glutathione (GSH) generated by the hexose monophosphate shunt [Watts, R.N. & Richardson, D.R. (2001) J. Biol. Chem. 276, 4724–4732]. We hypothesized that GSH completes the coordination shell of an NO–Fe complex that is released from the cell.
