Tissue culture was first devised at the beginning of the
twentieth century [Harrison, 1907; Carrel, 1912] (Table 1.1)
as a method for studying the behavior of animal cells free
of systemic variations that might arise in vivo both during
normal homeostasis and under the stress of an experiment.
As the name implies, the technique was elaborated first
with undisaggregated fragments of tissue, and growth was
restricted to the migration of cells from the tissue fragment,
with occasional mitoses in the outgrowth.
The purpose of this book is to provide the advances in plant in vitro culture as related to perennial fruit crops and medicinal plants. Basic principles and new techniques, now available, are presented in detail. The book will be of use to researchers, teachers in biotechnology and for individuals interested to the commercial application of plant in vitro culture.
Since the publication of this book, in 1983, several new and exciting
developments have taken place in the field of Plant Tissue Culture, and
it now forms a major component of what is popularly called Plant Biotechnology.
Many of the important crop plants which were then regarded
as recalcitrant are now amenable to regeneration from cultured protoplasts,
cells, and calli, enabling subjection of these crops to improvement
by biotechnological methods of cell manipulation.
Plant tissue culture
Most methods of plant transformation applied to GM crops require that a whole plant is regenerated from isolated plant cells or tissue which have been genetically transformed. This regeneration is conducted in vitro so that the environment and growth medium can be manipulated to ensure a high frequency of regeneration. In addition to a high frequency of regeneration, the regenerable cells must be accessible to gene transfer by whatever technique is chosen (gene transfer methods are described in Chapter 3).

At present, gamma irradiation has yet to be used in the processing of tissue
allografts by all tissue banks. The Massachusetts General Hospital Tissue
Bank inBoston—set up in 1990 by the pioneerDrHenry J.Mankin—is still
a surgical tissue bank employing sterile procurement and processing techniques,
but not gamma irradiation (Mankin was succeeded by Dr William
Tomford). Similarly, in Latin America, the Musculoskeletal Tissue Bank in
Latin Hospital, Buenos Aires, Argentina, set up by another famous pioneer
Dr Ottolenghi (now run by Dr Musculo), is also a surgical tissue bank.
