Chromatin is a highly dynamic structure that plays an essential role in regulating all nuclear processes that utilize the DNA template including DNA repair, replication, transcription and recombination. Thus, the mechanisms by which chromatin structures are assembled and modified are questions of broad interest. This minireview will focus on two groups of proteins: (a) histone chaperones and (b) ATPdependent chromatin remodeling machines, that co-operate to assemble DNA and histone proteins into chromatin....

The RuvABC resolvasome ofEscherichia colicatalyses the resolution of Holliday junctions that arise during genetic recombination and DNA repair. This process involves two key steps: branch migration, catalysed by the RuvB protein that is targeted to the Holliday junction by the structure specific RuvAprotein, and resolution, which is catalysed by the RuvC endonuclease.

The dermal glands of frogs produce antimicrobial peptides that protect the skin against noxious microorganisms and assist in wound repair. The sequences of these peptides are very dissimilar, both within and between species, so that the 5000 living anuran frogs may produce100 000 different antimicrobial peptides.

We have previously reported that sulfoquinovosylmonoacylglycerol (SQMG) is a potent inhibitor of mammalian DNA polymerases. DNA polymeraseb(pol b) is one of the most important enzymes protecting the cell against DNA damage by base excision repair. In this study, we charac-terized the inhibitory action of SQMG against rat pol b. SQMG competed with both the substrate and the template-primer for binding to polb.

Replication protein A (RPA), a heterotrimeric protein composed of 70, 32 and 14-kDa subunits, has been shown to be essential for DNA replication, repair, recombination, and transcription. Previously, we found that, in two seed plants, rice and Arabidopsis, there are two different types of RPA70-kDa subunit. Substantial biochemical and genetic characterization of these two subunits, termed OsRPA70aand OsRPA70bor AtRPA70a and AtRPA70b, respectively, is described in this report.

Spontaneous protein deamidation of labile Asn residues, generatingl-iso-aspartates andd-aspartates, is associated with cell aging and is enhanced by an oxidative microenvironment; to minimize the damage, the isoaspar-tate residues can be ‘repaired’ by a specific l-isoaspartate (d-aspartate) protein O-methyltransferase (PIMT).

Neuroinflammatory mediators play a crucial role in the pathophysiology of brain ischemia, exerting either deleterious effects on the progression of tis-sue damage or beneficial roles during recovery and repair. Within hours after the ischemic insult, increased levels of cytokines and chemokines enhance the expression of adhesion molecules on cerebral endothelial cells,

High molecular weight kininogen (HK) is an abundant plasma protein that plays a central role for the function of the kallikrein⁄kinin⁄kininogen sys-tem. Thus, cleavage of HK by kallikrein liberates bradykinin, which stimu-lates vascular repair and a two-chain protein, activated HK (HKa), which induces apoptosis in proliferating endothelial cells.

Homologous recombination is a very important cellular process, as it pro-vides a major pathway for the repair of DNA double-strand breaks. This complex process is affected by many factors within cells. Here, we have studied the effect of monovalent cations (K + ,Na + , and NH4 + ) on the outcome of recombination events, as their presence affects the biochemical activities of the proteins involved in recombination as well as the structure of DNA.

Protein kinases drive the cellular signal transduction networks that underlie the regulation of growth, survival and differentiation. To repair the deregu-lations of signaling cascades that are associated with numerous disease states, therapeutic strategies, based on controlling aberrant protein kinase activity, are emerging.

The term ‘antimicrobial peptides’ refers to a large number of peptides first characterized on the basis of their antibiotic and antifungal activities. In addition to their role as endogenous antibiotics, antimicrobial peptides, also called host defense peptides, participate in multiple aspects of immunity (inflammation, wound repair, and regulation of the adaptive immune sys-tem) as well as in maintaining homeostasis.

EVL is a member of the ENA⁄VASP family, which is involved in actin-remodeling processes. Previously, we reported that human EVL directly interacts with RAD51, which is an essential protein in the homologous recombinational repair of DNA double-strand breaks, and stimulates RAD51-mediated recombination reactionsin vitro.
