The Palpable Breast Mass Women should be strongly encouraged to examine their breasts monthly. A potentially flawed study from China has suggested that BSE does not alter survival, but given its safety, the procedure should still be encouraged. At worst, this practice increases the likelihood of detecting a mass at a smaller size when it can be treated with more limited surgery. Breast examination by the physician should be performed in good light so as to see retractions and other skin changes.

The composition of the chloroplast-localized protease complex, ClpP, from the green alga Chlamydomonas reinhardtiiwas characterized by nondena-turing electrophoresis, immunoblotting and MS. The detected ClpP complex has a native mass of540 kDa, which is200 kDa higher than ClpP complexes in higher plant chloroplasts, mitochondria or bacteria. The 540-kDa ClpP complex contains two nuclear-encoded ClpP proteins (ClpP3 and P5) and five ClpR (R1, R2, R3, R4 and R6) proteins, as well two pro-teins, ClpP1Land ClpP1H, both probably derived from the plastid clpP1 gene. ...

Synthetic hyaluronan oligosaccharides with defined structures and their pyridylaminated derivatives were used to investigate the mechanism of hydrolysis of hyaluronan by bovine testicular hyaluronidase. The products of the hydrolysis were analyzed by HPLC and ion-spray mass spectroscopy (MS).

Many branchiopod crustaceans are endowed with extracellular, high-molecular-weight hemoglobins whose exact structural characteristics have remained a matter of conjecture. By using a broad spectrum of techniques, we provide precise and coherent information on the hemoglobin of one of the phylogenetically ‘oldest’ extant branchiopods, the tadpole shrimp Triops cancriformis.

Neural stimulation controls the contractile properties of skeletal muscle fibres through transcriptional regulation of a number of proteins, including myosin isoforms. To study whether neural stimulation is also involved in the control of post-translational modifications of myosin, we analysed the phosphorylation of alkali myosin light chains (MLC1) and regulatory myo-sin light chains (MLC2) in rat slow (soleus) and fast (extensor digitorum longus EDL) muscles using 2D-gel electrophoresis and mass spectrometry....

The LmbB1 protein, participating in the biosynthesis of lincomycin, was heterologously expressed inEscherichia coli, purified in its active form, and characterized as a dimer of identical subunits. Methods for purification and analysis of the LmbB1 reaction product were developed. Molecular mass and fragmentation pattern of the product revealed by capillary electrophoresis-mass spectrometry were in agree-ment with its proposed structure, 4-(3-carboxy-3-oxo-propenyl)-2,3-dihydro-1H-pyrrole-2-carboxylic acid.

A cellulase [endo-b-1,4-D-glucanase (EC 3.2.1.4)] was iso-lated from the hepatopancreas of abalone Haliotis discus hannaiby successive chromatographies on TOYOPEARL CM-650M, hydroxyapatite and Sephacryl S-200 HR. The molecular mass of the cellulase was estimated to be 66 000 Da by SDS/PAGE, thus the enzyme was named HdEG66. The hydrolytic activity of HdEG66 toward carb-oxymethylcellulose showed optimal temperature and pH at 38Cand 6.3, respectively.

We report the biochemical characterization of calhepatin, a calcium-binding protein of the S100 family, isolated from lungfish (Lepidosiren paradoxa) liver. The primary structure, determined by Edman degradation and MS/MS, shows that the sequence identities with the other members of the family are lower than those between S100 proteins from different species. Calhepatin is composed of 75 residues and has a molecular mass of 8670 Da. It is smaller than calbindin D9k (78 residues), the smallest S100 described so far....

The propagation of yeast prion phenotypes is highly dependent on molecu-lar chaperones. We previously demonstrated that the molecular chaperone Ssa1p sequesters Ure2p in high molecular weight, assembly incompetent oligomeric species. We also determined the affinity of Ssa1p for Ure2p, and its globular domain.

Urine, amniotic fluid and ascitic fluid samples of galactosialidosis patients were analyzed and structurally characterized for free oligosaccharides using capillary high-performance anion-exchange chromatography with pulsed amperometric detection and online mass spectrometry. In addition to the expected endo-b-N-acetylglucosaminidase-cleaved products of complex-type sialylated N-glycans, O-sulfated oligosaccharide moieties were detected.

The extracellular bga1-encoded b-galactosidase of Hypocrea jecorina (Trichoderma reesei) was overexpressed under the pyruvat kinase (pki1) promoter region and purified to apparent homogeneity. The monomeric enzyme is a glycoprotein with a molecular mass of 118.8 ± 0.5 kDa (MALDI-MS) and an isoelectric point of 6.6.

In animal cells, many proteins have been shown to undergo glutathionyla-tion under conditions of oxidative stress. By contrast, very little is known about this post-translational modification in plants. In the present work, we showed, using mass spectrometry, that the recombinant chloroplast A4-glyceraldehyde-3-phosphate dehydrogenase (A4-GAPDH) from Arabid-opsis thaliana is glutathionylated with either oxidized glutathione or reduced glutathione and H2O2.

The protein assembly and stability of photosystem II (PSII) (sub)complexes were studied inmature leaves of four plastid mutants of tobacco (Nicotiana tabacumL), each havingone of thepsbEFLJoperon genes inactivated. In the absence of psbL, no PSII core dimers or PSII–light harvesting complex (LHCII) supercomplexes were formed, and the assembly of CP43 into PSII core monomers was extremely labile. The assembly of CP43 intoPSII coremonomers was found to be necessary for the assembly of PsbO on the lumenal side of PSII. ...

The presenilin proteins are required for intramembrane cleavage of a subset of type 1 membrane proteins including the Alzheimer’s disease amyloid precursor protein. Previous studies indicate presenilin proteins form enzymatically act-ive high molecular mass complexes consisting of hetero-dimers of N- and C-terminal fragments in association with nicastrin, presenilin enhancer-2 and anterior pharynx defective-1 proteins.

Phaiodactylipinwaspurified fromthevenomof the scorpion Anuroctonus phaiodactylus. It is the first protein to be puri-fied from a scorpion of the family Iuridae and has a molecular mass of 19 172 atomic mass units. The mature protein is composedof two subunits, the large one consisting of 108 amino acid residues, whereas the small subunit has only 18 residues, and the structure is stabilized by five disulfide bridges. The heterodimer is expressed froma single message containing 769 base pairs and a signal peptide with 16 and/or 25 amino acid residues. ...

A hallmark ofa-crystallin-type small heat shock proteins (sHsps) is their highly dynamic oligomeric structure which promotes intermolecular interactions involved in subunit exchange and substrate binding (chaperone-like activity). We studied the oligomeric features of two classes of bacterial sHsps by size exclusion chromatography and nanoelectro-spray mass spectrometry.Proteins of both classes formed large complexes that rapidly dissociated upon dilution and at physiologically relevant heat shock temperatures....
