In recent years major progress has been made in elucidating the mechanism and structure of catalytic RNA molecules, and we are now beginning to understand ribozymes well enough to turn them into useful tools. Work in our laboratory has focused on the development of twin ribozymes for site-specific RNA sequence alteration. To this end, we followed a strategy that relies on the combination of two ribozyme units into one molecule (hence dubbed twin ribozyme).

Laccase-catalyzed polymerization of tyrosine and tyrosine-containing pep-tides was studied in the presence and absence of ferulic acid (FA). Advanced spectroscopic methods such as MALDI-TOF MS, EPR, FTIR microscopy and HPLC-fluorescence, as well as more conventional analyt-ical tools: oxygen consumption measurements and SDS⁄PAGE were used in the reaction mechanism studies. Laccase was found to oxidize tyrosine and tyrosine-containing peptides, with consequent polymerization of the compounds.

With the amount of genetic information available, a lot of attention has focused on systems biology, in particular biomolecular interactions. Con-sidering the huge number of such interactions, and their often weak and transient nature, conventional experimental methods such as X-ray crystal-lography and NMR spectroscopy are not sufficient to gain structural insight into these. A wealth of biochemical and⁄or biophysical data can, however, readily be obtained for biomolecular complexes.

The cAMP response element consensus sequence directs the transcription of a wide range of genes. A 24-mer single-stranded cAMP response element decoy oligonucleotide (CDO) has been shown to compete with these sequences for binding transcription factors and therefore interferes with cAMP-induced gene transcription. We have examined the effect of this CDOalone and in combinationwith a range of common chemotherapeutic agents in colorectal cancer cell lines.

The expression of the colonic mitochondrial 3-hydroxy 3-methyl glutaryl CoA (mHMGCoA) synthase,a key con-trol site of ketogenesis from butyrate,is lower in germ-free (GF) than in conventional (CV) rats. In contrast,the activity of glutaminase is higher. The objective of this study was to investigate whether the intestinal flora can affect gene expression through short chain fatty acid (SCFA) and butyrate production.
