Polyhydroxyalkanoate (PHA) is stored as an important carbon and energy source in bacterial cells. For biomedical applications, gram-positive bacteria can be better sources of PHAs, since they lack outer membrane lipopolysaccharide. Although gram-positive Streptomyces coelicolor A3(2) has been indicated as a high potential PHA producer, phaC gene that encodes the key enzyme PHA synthase in the metabolic pathway is not determined in its genome.

We have investigated the crr gene of Streptomyces coelicolor that encodes a homologue of enzyme IIAGlucose of Escherichia coli, which, as a component of the phosphoenolpyruvate-dependent sugar phosphotransferase system (PTS) plays a key role in carbon regulation by triggering glucose transport, carbon catabolite repression, and inducer exclusion. As in E. coli, the crr gene of S. coelicolor is genetically associated with the ptsI gene that encodes the general phosphotransferase enzyme I.

The folding of a 93-residue protein, the histidine-phospho-carrier protein ofStreptomyces coelicolor,HPr,hasbeen studied using several biophysical techniques, namely fluo-rescence, 8-anilinonaphthalene-1-sulfate binding, circular dichroism, Fourier transform infrared spectroscopy, gel filtration chromatography and differential scanning calori-metry. The chemical-denaturation behaviour of HPr, fol-lowed by fluorescence, CD and gel filtration, at pH 7.5 and 25C, is described as a two-state process, which does not involve the accumulation of thermodynamically stable intermediates.

TheSCO2299gene fromStreptomyces coelicolorencodes a single peptide consisting of 497 amino acid residues. Its N-terminal region shows high amino acid sequence similarity to RNase HI, whereas its C-terminal region bears similarity to the CobC protein, which is involved in the synthesis of cobalamin. The SCO2299gene suppressed a temperature-sensitive growth defect of anEscherichia coliRNase H-deficient strain, and the recombinant SCO2299 protein cleaved an RNA strand of RNAÆDNA hybridin vitro.

In Gram-positive bacteria, the expression of iron-regulated genes is medi-ated by a class of divalent metal-dependent regulatory (DmdR) proteins. We cloned and characterized twodmdR genes of Streptomyces coelicolor that were located in two different nonoverlapping cosmids. Functional ana-lysis of dmdR1anddmdR2was performed by deletion of each copy. Dele-tion of dmdR1resulted in the derepression of at least eight proteins and in the repression of three others, as shown by 2D proteome analysis.

Thermodynamic parameters describing the conformational stability of the histidine-containing phosphocarrier protein fromStreptomyces coelicolor, scHPr, have been determined by steady-state fluorescence measurements of isothermal urea-denaturations, differential scanning calorimetry at different guanidinium hydrochloride concentrations and, independently, by far-UV circular dichroism measurements of isothermal urea-denaturations, and thermal denatura-tions at fixed urea concentrations.

ThedmdR1gene ofStreptomyces coelicolorencodes an important regulator of iron metabolism. An antiparallel gene (adm) homologous to a develop-ment-regulated gene ofStreptomyces aureofacienshas been found to over-lap with dmdR1. Both proteins DmdR1 and Adm are formed in solid and liquid cultures of S. coelicolorA3(2).
