Streptozotocin

In diabetic patients, the body does not produce enough insulin or not properly respond to insulin. Insulin is a hormone produced by -cells of the pancreas. It stimulates the cells to absorb the glucose from the blood and its hyposecretion may leads to the over accumulation of glucose in the blood and often leading to various complications.

Liver metabolism is influenced by hormones and nutrients. Amino acids such as glutamine or leucine induce an anabolic response, which resembles that of insulin in muscle and adipose tissue. In this work, the signalling pathways and the effects of insulin were compared to those of glutamine and leucine in isolated hepatocytes from normal and streptozotocin-diabetic rats.

The levels ofa-aminoadipic-d-semialdehyde residue, the oxidative deamination product of lysine residue, in plasma protein from streptozotocin-induced diabetic rats were evaluated.a-Aminoadipic-d-semialdehyde was converted to a bisphenol derivative by acid hydrolysis in the presence of phenol, and determined by high performance liquid chro-matography. Analysis of plasma proteins revealed three timeshigherlevelsofa-aminoadipic-d-semialdehyde in dia-betic subjects comparedwithnormal controls.

Oxidative stress, followed by the apoptotic death of pancreaticbcells, is considered to be one of causative agents in the evolution of the type 2 dia-betic state; therefore, the protection ofbcells can comprise an efficacious strategy for preventing type 2 diabetes. In the present study, RIN-m5F cells (i.e. the rat insulinoma bcell line) were stimulated with streptozotocin, resulting in a time- and concentration-dependent release of lactate dehydrogenase.

The ubiquitous isoform of 6-phosphofructo-2-kinase⁄fructose-2,6-bisphos-phatase (uPFK-2), a product of thePfkfb3gene, plays a crucial role in the control of glycolytic flux. In this study, we demonstrate that Pfkfb3gene expression is increased in streptozotocin-induced diabetic mouse liver. The Pfkfb3⁄-3566 promoter construct linked to the luciferase reporter gene was delivered to the liver via hydrodynamic gene transfer.

Abstract Curcumin, an active principle component in rhizome of Curcuma longa, has proved its merit for diabetes through its anti-oxidative and anti-inflammatory properties. This study aims at evaluating the effect of curcumin in modulating the altered dopaminergic receptors, CREB and phospholipase C in the cerebral cortex and cerebellum of STZ induced diabetic rats. Radioreceptor binding assays and gene expression was done in the cerebral cortex and cerebellum of male Wistar rats using specific ligands and probes.
