This study determines the interaction between RBPs and target-RNA complexes from public data of the ENCODE project and identifies mutations associated with Mendelian diseases that could disrupt the RBP-RNA interactions.

Murine double minute 2 (MDM2) protein exhibits many diverse biochemi-cal functions on the tumour suppressor protein p53, including transcrip-tional suppression and E3 ubiquitin ligase activity. However, more recent data have shown that MDM2 can exhibit ATP-dependent molecular chap-erone activity and directly mediate folding of the p53 tetramer. Analysing the ATP-dependent function of MDM2 will provide novel insights into the evolution and function of the protein.

The interaction between the c11ring and thececomplex, forming the rotor of theIlyobacter tartaricus ATP synthase, was probed by surface plasmon resonance spectroscopy andin vitro reconstitution analysis. The results pro-vide, for the first time, a direct and quantitative assessment of the stability of the rotor. The data indicated very tight binding between the c11ring and the cecomplex, with an apparent Kd value of approximately 7.4 nm.

A computer program has been evaluated for subsite map calculations of depolymerases. The program runs inWIN-DOWSanduses the experimentallydeterminedbond cleavage frequencies (BCFs) for determination of the number of subsites, the position of the catalytic site and for calculation of subsite binding energies. The apparent free energy values were optimized by minimization of the differences of the measured and calculated BCF data.

The Ca 2+ -induced transition in the troponin complex (Tn) regulates vertebrate striated muscle contraction. Tn was reconstituted with recombinant forms of troponin I (TnI) containing a single intrinsic 5-hydroxytryptophan (5HW). Fluorescence analysis of these mutants of TnI demonstrate that the regions in TnI that respond to Ca 2+ binding to the regulatoryN-domain of TnC are the inhibitoryregion (residues 96–116) and a neighboring region that includes position 121.

With the aim of extending our knowledge on the reaction pathways of Zn-metallothionein (MT) and apo-MT species in the presence of Hg(II), we monitored the titration of Zn7-MT, Zn4 -aMT and Zn3 -bMT proteins, at pH 7 and 3, with either HgCl2or Hg(ClO4 )2 by CD and UV-vis spectr-oscopy.
