In this thesis, the author focussed on providing an application framework to be used in brain segmentation - the process of tumor detection. This constitutes setting up software bundled into a Window machine which is easily distributable. Moreover, a segmentation algorithm was implemented and tested using the provided software. In order to test the bundled software by implementing specific brain segmentation from the literature, the author used CT scans freely available from the database of 108 Central Military Hospital.

The thesis is the first study which evaluates the results of formultiple organs resuscitation strategy in brain death potential donors. From the results of this study, it is recommended to optimize the hemodynamics to adequately perfusion of organs after brain death to ensure the functions of the transplanted organs later; the organs should be used in the 24 first hours frombeginning of organ resuscitation (about 12 hours after the declaring of Brain-dead diagnosis Council) because the organ functionsare is the best.

Wilson disease (WD), an inherited disorder affecting copper metabolism, is characterized by hepatic cirrhosis and neuronal degeneration, which result from toxic levels of copper that accumulate in the liver and brain, respectively. We reported previously that the 1.3-kb promoter of the WD gene contains four metal response elements (MREs). Among the four MREs, MREa plays the most important role in the transcriptional activation of the WD promoter.

BLASTanalysis of the human and mouse genome sequence databases using the sequence of the human CMP-sialic acid:b-galactoside a-2,6-sialyltransferase cDNA (hST6Gal I, EC2.4.99.1) as a probe allowed us to identify a putative sialyltransferase gene on chromosome 2. The sequence of the corresponding cDNA was also found as an expressed sequence tag of human brain.

We isolated two novel 14-3-3 binding proteins using 14-3-3fas bait in a yeast two-hybrid screen of a human brain cDNA library. One of these encoded the C-terminus of a neural specific armadillo-repeat protein, d-catenin (neural plakophilin-related arm-repeat protein or neurojungin). d-Catenin from brain lysates was retained on a 14-3-3 affinity column.

RasGRF is a family of guanine nucleotide exchange factors with dual spe-cificity for both Ras and Rac GTPases. In this study, using mouse brain extracts, we show that both RasGRF1 and RasGRF2 interact with micro-tubules in an in vitro microtubule assembly system and this binding is very tight. To characterize this association, recombinant purified proteins con-taining different regions of RasGRF1 were tested for their ability to bind microtubules preassembled from pure tubulin.

Several molecular changes occur following axotomy, such as gene up-regulation and down-regulation. In our previous study using Affymetrix arrays, it was found that after the axotomy of sciatic nerve, there were many novel genes with significant expression changes. Among them, neuronatin (Nnat) was the one which expression was significantly up-regulated. Nnat was identified as a gene selectively expressed in neonatal brains and markedly reduced in adult brains.
