In this Masters thesis computational modelling techniques were employed to investigate iron-free apo-Lactoferrin (apo-Lf) structural conformation changes in the presence of variant temperature and pH. These conditions represent the environment most milk protein goes through in food processing and the production of food products.

The development of fruits without fertilization is known as parthenocarpy. Parthenocarpy improves the fruit quality, processing attributes, production and productivity of vegetable crops like tomato, cucumber, watermelon etc. Absence of seeds can enhances the shelf life of the fruits, allowing a better conservation, fruit set in adverse climatic conditions, early and offseason production of vegetable crops. Therefore, it is important to ensure yield stability regardless of environmental conditions.

The catalyst 7.5% NiO/TiO2 and seven samples of NiO/γ-Al2O3 with NiO content from 7.5% to 60% have been obtained and studied. Physico-chemical characteristics of the catalysts were determined by methods of Adsorption (BET), X-ray Diffraction (XRD), TemperatureProgrammed Reduction (TPR) and Hydrogen Pulse Titration. The catalytic properties of the obtained samples were investigated in the reaction of CO methanation at temperatures 180 – 220◦C and mole ratios hydrogen/carbon monoxide 25-100.

It is scientifically and industrially important 1 to clarify the stabilizing mechanism of proteases in extraordinary envi-ronments. We used subtilisins ALPI and Sendai as models to study the mechanism. Subtilisin ALPI is extremely sensitive to highly alkaline conditions, even though the enzyme is produced by alkalophilicBacillus, whereas sub-tilisin Sendai from alkalophilic Bacillusis stable under conditions of high alkalinity.

Escherichia coliproteinY (pY) binds to the small ribosomal subunit and stabilizes ribosomes against dissociation when bacteria experience environmental stress. pY inhibits trans-lationin vitro, most probably by interferingwith the binding of the aminoacyl-tRNA to the ribosomal A site. Such a translational arrest may mediate overall adaptation of cells to environmental conditions. We have determined the 3D solution structure of a 112-residue pY and have studied its backbone dynamic by NMRspectroscopy. ...

UMP-CMP kinase catalyses an important step in the phosphorylationofUTP, CTPanddCTP. It is also involved in the necessary phosphorylation by cellular kinases of nucleoside analogs used in antiviral therapies. The reactivity of human UMP-CMP kinase towards natural substrates and nucleotide analogs was reexamined. The expression of the recombinant enzyme and conditions for stability of the enzyme were improved. Substrate inhibition was observed for UMP and CMP at concentrations higher than 0.2 mM, but not for dCMP.

Using 0.4Mimidazole citrate buffer (pH 7.5) containing 0.1 mML-cysteine, homodimeric starch phosphorylase from Corynebacterium calluane(CcStP) was dissociated into native-like folded subunits concomitant with release of pyridoxal 5¢-phosphate and loss of activity. The inactivation rate of CcStP under resolution conditions at 30Cwas, respectively, four- and threefold reduced in two mutants, Arg234fiAla and Arg242fiAla, previously shown to cause thermostabilization of CcStP [Griessler, R., Schwarz, A., Mucha, J.&Nidetzky, B. ...

We have studied the stability of the histone-like, DNA-binding protein HU from the hyperthermophilic eubacteriumThermotoga maritimaand its E34D mutant by differential scanningmicrocalorimetry and CDunder acidic conditions at various concentrations within the range of 2–225lMof monomer. The thermal unfolding of both proteins is highly reversible and clearly follows a two-state dissociation/unfolding model from the folded, dimeric state to the unfolded, monomericone.

Life grows almost everywhere on earth, including in extreme environments and under harsh conditions. Organisms adapted to high temperatures are called thermophiles (growth temperature 45–75C) and hyperthermophiles (growth temperature ‡80C). Proteins from such organisms usually show extreme thermal stability, despite having folded structures very similar to their mesostable counterparts.

Neuroglobin (Ngb) is a recently discovered globin that is predominantly expressed in the brain, retina and other nerve tissues of human and other vertebrates. Ngb has been shown to act as a neuroprotective factor, pro-moting neuronal survival in conditions of hypoxic–ischemic insult, such as those occurring during stroke.

In ruminants, some leaf-eating animals, and some insects, defensive lyso-zymes have been adapted to become digestive enzymes, in order to digest bacteria in the stomach. Digestive lysozyme has been reported to be resis-tant to protease and to have optimal activity at acidic pH. The structural basis of the adaptation providing persistence of lytic activity under severe gastric conditions remains unclear.
