Amlodipine is a representative active ingredient of calcium channel blockers. In addition to the general advantages of calcium channel blockers in treating hypertension, amlodipine also effectively controls blood pressure for 24 hours and has no or just a little effect on neurohormonal activation, so it does not cause high blood pressure at the last dose. However, amlodipine has a plasma half-life of between 30 and 40 hours, slowing the drug's action duration.

Valsartan, an angiotensin blocker, is often combined with amlodipine (a calcium channel antagonist) in the current treatment of hypertension. There is a generic brand-name drug Exforge® which has been proven to be clinically effective. However, its price is high, making it quite difficult to access, especially in low- and middle-income countries. Therefore, research and development of generic drugs is essential.

To evaluate the effect of the drugs in terms of ambulatory BP (ABPM) and clinical reductions, as well as analysing their effect on some variables such as BP morning surge, trough-peak ratio and smoothness index, which have been shown to be predictive of cardiovascular and cerebrovascular risk.

The combination of hydrochlorothiazide (HCT), amlodipine besylate (AML) and olmesartan medoxomil (OLM) is highly effective in treating hypertension. Currently, Vietnamese Pharmacopoeia V and other reference pharmacopoeias do not have a procedure for the simultaneous quantification of these three ingredients. To develop and validate a procedure for simultaneous determination of HCT, AML and OLM in tablets by HPLC method.
