Báo cáo khoa học: The miRNA-192 ⁄194 cluster regulates the Period gene family and the circadian clock
Several biological functions in mammals are regulated in a circadian fash-ion. The molecular mechanisms orchestrating these circadian rhythms have been unravelled. The biological clock, with its core transcriptional unit Bmal1⁄CLOCK, is composed of several self-sustaining feedback loops. In this study, we describe another mechanism impinging on the core compo-nents of the circadian clock.

Báo cáo khoa học: Time-dependent regulation analysis dissects shifts between metabolic and gene-expression regulation during nitrogen starvation in baker’s yeast
Time-dependent regulation analysis is a new methodology that allows us to unravel, both quantitatively and dynamically, how and when functional changes in the cell are brought about by the interplay of gene expression and metabolism. In this first experimental implementation, we dissect the initial and late response of baker’s yeast upon a switch from glucose-lim-ited growth to nitrogen starvation.

Báo cáo khoa học: MicroRNA-9 inhibits ovarian cancer cell growth through regulation of NF-jB1
MicroRNAs are emerging as important regulators of cancer-related pro-cesses. Our studies show that microRNA-9 (miR-9) is downregulated in human ovarian cancer relative to normal ovary, and overexpression of miR-9 suppresses cell growthin vitro. Furthermore, the 3¢-UTR of NF-jB1 mRNA is found to be regulated directly by miR-9, demonstrating that NF-jB1 is a functionally important target of miR-9 in ovarian cancer cells.

Báo cáo khoa học: Evolutionary changes to transthyretin: developmentally regulated and tissue-specific gene expression
A survey of the expression of the transthyretin and thyroxine-binding glob-ulin genes in various species during development provides clues as to how the present thyroid hormone distribution network in extracellular compart-ments developed during vertebrate evolution. Albumin may be the ‘oldest’ component of the thyroid hormone distribution network as it is found in the plasma of all vertebrates investigated.

Báo cáo khoa học: Dynamic interactions of proteins in complex networks: a more structured view
Virtually every process in a cell is carried out by macromolecular com-plexes whose actions need to be perfectly orchestrated. The synchronization and regulation of these biological functions is indeed critical and is usually carried out by complex networks of transient protein interactions. Here, we review some of the many strategies that proteins in regulatory networks use to achieve the dynamic plasticity necessary to rapidly respond to diverse cellular needs.

Báo cáo khoa học: Dehydroepiandrosterone inhibits the proliferation and induces the death of HPV-positive and HPV-negative cervical cancer cells through an androgen- and estrogen-receptor independent mechanism
Dehydroepiandrosterone (DHEA) has a protective role against epithelial-derived carcinomas; however, the mechanisms remain unknown. We deter-mined the effect of DHEA on cell proliferation, the cell cycle and cell death in three cell lines derived from human uterine cervical cancers infected or not with human papilloma virus (HPV).

Báo cáo khoa học: Suppression of nuclear factor-jB activity in macrophages by chylomicron remnants: modulation by the fatty acid composition of the particles
Current evidence indicates that chylomicron remnants (CMR) induce macrophage foam cell formation, an early event in atherosclerosis. Inflam-mation also plays a part in atherogenesis and the transcription factor nuclear factor-jB (NF-jB) has been implicated. In this study, the influence of CMR on the activity of NF-jB in macrophages and its modulation by the fatty acid composition of the particles were investigated using macro-phages derived from the human monocyte cell line THP-1 and CMR-like particles (CRLPs)....

Báo cáo khoa học: An orphan dermaseptin from frog skin reversibly assembles to amyloid-like aggregates in a pH-dependent fashion
Dermaseptin PD-3-7 (aDrs) from frog skin contains three aspartic acid resi-dues resulting in a negative net charge at neutral pH, as opposed to numer-ous other dermaseptins which are cationic helical antimicrobial peptides. Still, this peptide can be fitted into an amphipathicahelix by an Edmundson wheel projection.

Báo cáo khoa học: A moss pentatricopeptide repeat protein binds to the 3¢ end of plastid clpP pre-mRNA and assists with mRNA maturation
Pentatricopeptide repeat (PPR) proteins constitute a large family in land plants and are required for various post-transcriptional steps associated with RNA in plant organelles. The mossPhyscomitrella patensPPR protein, PpPPR_38, is a nuclear-encoded chloroplast protein and was previously shown to be involved in the maturation step of chloroplastclpPpre-mRNA.

Báo cáo khoa học: Analysis of the region for receptor binding and triggering of oligomerization on Bacillus thuringiensis Cry1Aa toxin
The determination of the receptor-binding region of Cry toxins produced byBacillus thuringiensisis expected to facilitate an improvement in their insecticidal ability through protein engineering. We analyzed the region on Cry1Aa molecules involved in interactions with the cadherin-like protein receptor BtR175 using cysteine-substituted mutant toxins and several syn-thetic peptides corresponding to the loops in domain 2.

Báo cáo khoa học: The Pseudomonas aeruginosa nirE gene encodes the S-adenosyl-L-methionine-dependent uroporphyrinogen III methyltransferase required for heme d1 biosynthesis
Biosynthesis of hemed1, the essential prosthetic group of the dissimilatory nitrite reductase cytochromecd1 , requires the methylation of the tetrapyr-role precursor uroporphyrinogen III at positions C-2 and C-7. We pro-duced Pseudomonas aeruginosaNirE, a putativeS-adenosyl-l-methionine (SAM)-dependent uroporphyrinogen III methyltransferase, as a recombi-nant protein inEscherichia coliand purified it to apparent homogeneity by metal chelate and gel filtration chromatography.

Báo cáo khoa học: The thioflavin T fluorescence assay for amyloid fibril detection can be biased by the presence of exogenous compounds
Thioflavin T (ThT) dye fluorescence is used regularly to quantify the for-mation and inhibition of amyloid fibrils in the presence of anti-amyloido-genic compounds such as polyphenols. However, in this study, it was shown, using three polyphenolics (curcumin, quercetin and resveratrol), that ThT fluorescence should be used with caution in the presence of such exogenous compounds.

Báo cáo khoa học: Donor and acceptor substrate selectivity among plant glycoside hydrolase family 32 enzymes
Plant family 32 glycoside hydrolase enzymes include hydrolases (cell wall invertases, fructan exohydrolases, vacuolar invertases) and fructosyltrans-ferases. These enzymes are very similar at the molecular and structural levels but are functionally different. Understanding the basis of the func-tional diversity in this family is a challenging task.
