Preparation of astaxanthin/pycnogenol nanoparticles: A delivery system with improved astaxanthin stability and bioavailability
Astaxanthin (AST) and pycnogenol (PYC) are natural bioactive compounds commonly used in pharmaceuticals and functional foods due to their highly antioxidant activity and potential health benefits. However, the widespread application of astaxanthin is hindered by its poor water solubility and highly unsaturated structure. In this study, nanoparticles co-delivering astaxanthin and pycnogenol (NAP) were fabricated to enhance the dispersibility, stability, and bioavailability of astaxanthin.

Báo cáo Y học: Two GPX-like proteins from Lycopersicon esculentum and Helianthus annuus are antioxidant enzymes with phospholipid hydroperoxide glutathione peroxidase and thioredoxin peroxidase activities
This study investigated the enzymatic function of two putative plant GPXs, GPXle1 from Lycopersicon esculentum and GPXha2 from Helianthus annuus, which show sequence identities with the mammalian phospholipid hydroperoxide glutathione peroxidase (PHGPX). Both purified recombinant proteins expressed in Escherichia coli show PHGPX activity by reducing alkyl, fatty acid and phospholipid hydroperoxides but not hydrogen peroxide in the presence of glutathione.

Báo cáo khoa học: Antioxidant protein 2 prevents methemoglobin formation in erythrocyte hemolysates
Antioxidant protein 2 (AOP2) is a member of a family of thiol-specific antioxidants, recently renamedperoxiredoxins, that evolvedas part of anelaborate systemtocounteract and control detrimental effects of oxygen radicals. AOP2 is found in endothelial cells, erythrocytes, monocytes, T andB cells, but not in granulocytes. AOP2 was found solely in the cytoplasm and was not associated with the nuclear or membrane fractions; neither was it detectable in plasma. Further experiments focused on the function of AOP2 in erythrocytes where it is closely associated with the hemo-globin complex, particularlywith theheme....

Báo cáo khoa học: A selenium-containing single-chain abzyme with potent antioxidant activity
Reactive oxygen species (ROS) are products of normal metabolic activities and are thought to be the cause of many diseases. A selenium-containing single-chain abzyme 2F3 (Se-2F3-scFv) that imitates glutathione peroxidase has been produced which has the capacity to remove ROS. To evaluate the antioxidant ability of Se-2F3-scFv, we con-structeda ferrous sulfate/ascorbate (Vc/Fe 2+ )-inducedmito-chondrial damage model system and investigated the capacity of Se-2F3-scFv to protect mitochondria from oxi-dative damage. Se-2F3-scFv markedly decreased mito-chondrial swelling, inhibited lipid peroxidation, and maintained the activity of cytochromecoxidase, in com-parison with Ebselen, a well-studied glutathione peroxidase mimic, indicating that Se-2F3-scFvhas potential for treating diseases mediated by ROS....

Báo cáo khoa học: Mixed lineage kinase LZK and antioxidant protein-1 activate NF-jB synergistically
Leucine zipper-bearing kinase (LZK) is a novel member of themixed lineage kinase (MLK) family [Sakuma, H., Ikeda, A.,Oka,S.,Kozutsumi,Y.,Zanetta,J.P., andKawasaki, T. (1997)J. Biol.Chem.272, 28622–28629].Wehavepreviously shown that LZK activates the c-Jun-NH2terminal kinase (JNK) pathway, but not the extracellular signal-related kinase (ERK) pathway, by acting as a mitogen-activated protein kinase kinase kinase (MAPKKK) [Ikeda, A., Hasegawa, K., Masaki, M., Moriguchi, T., Nishida, E., Kozutsumi, Y., Oka, S., and Kawasaki, T. Báo cáo khoa học: Antioxidant defences in cybrids harboring mtDNA mutations associated with Leber’s hereditary optic neuropathy
Oxidative stress and imbalance between free radical generation and detoxi-fication may play a pivotal role in the pathogenesis of Leber’s hereditary optic neuropathy (LHON). Mitochondria, carrying the homoplasmic 11778⁄ND4, 3460⁄ND1 and 14484⁄ND6 mtDNA point mutations associ-ated with LHON, were used to generate osteosarcoma-derived cybrids. Enhanced mitochondrial production of reactive oxygen species has recently been demonstrated in these cybrids [Beretta S, Mattavelli L, Sala G, Trem-olizzo L, Schapira AHV, Martinuzzi A, Carelli V & Ferrarese C (2004) Brain127, 2183–2192]. ...
