The interactions between photosystem I and five charge mutants of plastocyanin from the cyanobacteriumPhormi-dium laminosumwere investigatedin vitro. The dependence of the overall rate constant of reaction,k2, on ionic strength was investigated using laser flash photolysis. The rate con-stant of the wild-type reaction increased with ionic strength, indicating repulsion between the reaction partners. Remov-inganegativechargeonplastocyanin(D44A) acceleratedthe reactionandmade it independentof ionic strength; removing a positive charge adjacent to D44 (K53A) had little effect....

Protein folding can be modulatedin vivo by many factors. While chaperones act as folding catalysts and show broad substrate specificity, some pro-peptides specifically facilitate the folding of the mature protein to which they are bound. Potato carboxypeptidase inhibitor (PCI), a 39-residue pro-tein carboxypeptidase inhibitor, is synthesized in vivo as a precursor protein that includes a 27-residue N-terminal and a seven-residue C-terminal pro-regions.

We have studied the reaction native«denatured for the 33-kDa protein isolated from photosystem II. Sucrose and glycerol have profound effects on pressure-induced unfolding. The additives shift the equilibrium to the left; they also cause a significant decrease in the standard volume change (DV). The change in DVwas related to the sucrose and glycerol concentrations.

The xylanase inhibitor protein I (XIP-I), recently identified in wheat, inhib-its xylanases belonging to glycoside hydrolase families 10 (GH10) and 11 (GH11). Sequence and structural similarities indicate that XIP-I is related to chitinases of family GH18, despite its lack of enzymatic activity. Here we report the identification and biochemical characterization of a XIP-type inhibitor from rice.

The 1,3(4)-b-d-glucanases of glycoside hydrolase family 16 provide useful examples of versatile yet specific protein–carbohydrate interactions. In the present study, we report the X-ray structures of the 1,3(4)-b-d-glucanase Phanerochaete chrysosporiumLaminarinase 16A in complex withb-glucan products from laminarin (1.6 A˚ ) and lichenin (1.1 A ˚ ) hydrolysis. The G6G3G3G glucan, in complex with the enzyme, showed ab-1,6 branch in the acceptor site.

The milk protein proteose peptone component 3 (PP3), also called lacto-phorin, is a small phosphoglycoprotein that is expressed exclusively in lactating mammary tissue. The C-terminal part of the protein contains an amphipathic helix, which, upon proteolytic liberation, shows antibacterial activity. Previous studies indicate that PP3 forms multimeric structures and inhibits lipolysis in milk.
