Azopirillum brasilense is a plant-growth promoting nitrogen-fixing bacteria that is used as bio-fertilizer in agriculture. Since nitrogen fixation has a high-energy demand, the reduction of N2 to NH4 + by nitrogenase occurs only under limiting conditions of NH4 + and O2.

An experiment was carried out to isolate, characterize and screening of Azospirillum from tuberose rhizoplane. 40 bacterial strains were isolated showing characteristics of Azospirillum. Isolates were characterized based on utilization of glucose, biotin requirement, acid production in glucose peptone broth and denitrification test. Out of 40 isolates, 20 isolates required biotin for their growth and remaining 20 showed growth without biotin. 16 isolates showed positive for denitrification and they were tentatively identified as Azospirillum brasilense.

Co-inoculation of Arbuscular mycorrhizal fungi (AMF) and certain species of Plant Growth Promoting Rhizobacteria (PGPR) help to bring the poorly soluble nutrients into the soil solution, thus enhancing nutrient uptake. The present investigation was carried out with objectives to study the effect of co-inoculation of AMF and PGPR (viz. Burkholderia cepacia and Azospirillum brasilense) on soil Zn fractions and Zn uptake by maize. A field experiment was conducted in Randomized Block Design with 9 treatments, each replicated thrice.

The nursery experiment was conducted to observe influence of microbial fertilizers on growth of oil palm seedlings for two consecutive years. Azotobacter chroococcum, Azospirillum brasilense, Bacillus megaterium, Frateuria aurantia and Glomus aggregatum were the microbial fertilizers used individually, combindly and integrated with chemical fertilizers. All the treatments were found markedly superior to the control with respect to morphological and growth characters of oil palm seedlings.

The effect of different kinds of organic nutrients on biochemical composition of mulberry and soil status of S-36 mulberry garden was assessed. The pooled data of two crops revealed that mulberry raised with 100 % Recommended N through 20% each of Compost + Glyricidia maculate + Castorcake + Vermicompost and Urea + 10 kg each of Azospirillum brasilense + Aspergillus awamori Bio-fertilizer + remaining P, K through chemical fertilizers + Rec. FYM (T8) recorded significantly higher chemical composition of leaf viz., leaf nitrogen (3.31%), phosphorous (1.69 %), potassium (1.39%), calcium(2.

The exclusive use of plant juices, not as a mere supplement to synthetic culture media, for culturing rhizospheric microorganisms (RMO) is introduced here. Juices were prepared from desert (Mesembryanthemum crystallinum L., Zygophyllum album L., Carpobrotus edulis L.) as well as cultivated (Trifolium alexandrinum L., Beta vulgaris L.) plants. Colonies of RMO (Azospirillum brasilense, Enterobacter agglomerans and Klebsiella pneumoniae) nicely developed on surfaceinoculated agar plates prepared from crude and diluted juice of M. crystallinum (ice plant).

Analyses of the spatial localization and the functions of bacteria in host plant habitats through in situ identification by immunological and molecular genetic techniques combined with high resolving microscopic tools and 3D-image analysis contributed substantially to a better understanding of the functional interplay of the microbiota in plants. Among the molecular genetic methods, 16S-rRNA genes were of central importance to reconstruct the phylogeny of newly isolated bacteria and to localize them in situ.
