Banana micropropagation

Tieu Hong banana variety (Musa acuminata) is one of the most popular bananas in Vietnam because they have not only beautiful colour, delicious taste but also high nutrition, and significantly economical value. Nowadays, this banana variety is primarily propagated in wide-scale by micropropagation which is effective with a high propagation coefficient, thus, that can meet the increasing demand of people to enjoy it.

Four antagonistic microorganisms Trichoderma viride, Metarhizium anisopliae, Pseudomonas fluorescens and Bacillus thuringiensis and their consortia were used to suppress Colletotrichum musae (Berk and Curtis), the causal agent of anthracnose disease of micropropagated banana during 2014-16.

Genetic uniformity of in vitro raised plants is a prerequisite for production of quality planting material of banana. Embryogenic cultures were induced from immature male flower buds of two important cultivars of banana, ‘Rasthali’ (Silk-Musa AAB) and ‘Grand Naine’ (Cavendish-Musa AAA). Cell suspensions were established and regenerated plants were evaluated for their genetic stability using 10 inter simple sequence repeat markers.

Photomixotrophic technique refers to cell employment of endogenous and exogenous carbohydrate sources to obtain energy in the presence of CO2 enrichment. This investigation aimed to evaluate the photomixotrophic technique as a combination between various concentrations of CO2 enrichment, and several types of carbohydrate sources (sucrose, fructose, glucose) at different concentrations on growth parameters of banana among all micropropagation stages.

The cultivar Robusta forms the mainstay of commercial banana cultivation in India. An efficient micropropagation system with high multiplication rate will boost banana cultivation and assist banana based industries in the country. The shoot apices of banana cv. Robusta were cultured on two basal media MS and B5 supplemented with different concentrations and combinations of plant growth regulators (PGR). The tissue culture responses were observed for shoot multiplication and effect of subculture on rate of shoot multiplication till 8th subculture.

In this study, RAPD fingerprinting has been carried out for variants selected from orchards growing with micropropagated Robusta banana (Musa spp. ‘AAA’) plants. After isolation of quality DNA and preliminary screening of random primers, fourteen arbitrary (10-mers) primers showing intense unambiguous and reproducible PCR amplification have been selected for study. Among them OPA-19 and OPC-03 showed only monomorphic bands while OPA-02, OPA-09, OPA-13, OPA-14, OPB-6, OPB-15, OPC-01, OPD-10, OPF-04 and OPF-12 primers showed polymorphic bands.

The investigation entitled “Standardization of suitable treatment for sucker production of Malbhog (AAB) banana through macropropagation” was carried out at Instructional cum Research Farm, Department of Horticulture, Biswanath College of Agriculture, Assam Agricultural University, Biswanath Chariali with a view to standardize the best treatment combination suitable for production of suckers of Malbhg (AAB) banana through micropropagation.

The present study was carried out to monitor somaclonal variations of banana cv. Ney Poovan (AB) during micropropagation using random amplified polymorphic DNA (RAPD). Sword suckers were used as explants and regenerated on MS media supplemented with BAP alone, BAP + Kinetin and TDZ at varying levels of concentrations. The regenerated shoots were again subcultured on MS medium fortified with plant growth regulators at different concentrations. Rooting was obtained on ½ MS medium supplemented with 2.00 mg/l IBA and 2 g/l of activated charcoal.
