Methicillin-resistant Staphylococcus aureus (MRSA) is one of the most predominant agents that cause nosocomial infections. Objectives: To determine the rate of MRSA and the molecular characteristics of the coagulase encoding gene of these isolates based on polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP).

The authors develop a system of differential equations describing the motion of the structural system equipped with DFP bearings. The Runge-Kutta numerical method, specifically the ode15s function in MATLAB, was used to solve these equations. The results of the calculations provide a comparison of the damping efficiency of the structural system with and without the use of DFP bearings, assessing the effectiveness of vibration isolation during an earthquake, with three components X, Y, and vertical considered.

In the course of current research the procedure of obtaining of the piecewise linear vibration systems time and frequency responses have been developed. The method is partly analytical and partly numerical and is based on combining of linear solutions of different sections in a cycle of motion. In a row with the method of obtaining of the exact steady state time and frequency response, also it is provided sensitivity analysis of the vibration isolator.

Many groups of proteins play important roles in the cell’s response to various stresses. The molecular chaperone GroEL of Escherichia coli represents one such highly conserved family of stress proteins. We have observed that isolated GroEL complexes from stationary cultures contain various polypeptides that can be released from the chaperonin by GroES and/or ATP, and identified two such polypeptides as the proteins GatY and UP12.

A series of trehalose-based oligosaccharides were isolated from the cytoplasmic fraction of Mycobacterium smegmatis and purified by gel-filtration and paper chromatography and TLC. Their structures were determined by HPLC and GLC to determine sugar composition and ratios, MALDI-TOF MS to measure molecular mass, methylation analysis to determine linkages, 1H-NMR to obtain anomeric configurations of glycosidic linkages, and exoglycosidase digestions followed by TLC to determine sequences and anomeric configurations of the monosaccharides....

For the biosynthesis of punicic acid (18:3 D9Z,11E,13Z )a (11,14)-linoleoyl desaturase activity has been proposed. To isolate this acyl-lipid-desaturase, PCR-based cloning was used. This approach resulted in the isolationof twocomplete cDNAs. The first isolated full-length cDNA harbors a sequence of 1350 bp encoding a protein of 395 amino acids. The second cDNA was 1415 bp long encoding a protein of 387 amino acids.

The photosynthetic protein complexes in plants are located in the chloroplast thylakoid membranes. These membranes have an ultrastructure that consists of tightly stackedÔgranaÕ regions interconnectedbyunstackedmembrane regions. The structure of isolated granamembranes has been studiedhere by cryo-electron microscopy. The data reveals an unusual arrangement of the photosynthetic protein complexes, staggered over two tightly stacked planes.

Pi4 is a38-residue toxincross-linkedby fourdisulfidebridges that has been isolated from the venom of the Chactidae scorpion Pandinus imperator. Together with maurotoxin, Pi1, Pi7 andHsTx1, Pi4belongs to theaKTX6 subfamilyof short four-disulfide-bridged scorpion toxins acting on K + channels. Due to its very low abundance in venom, Pi4 was chemically synthesized in order to better characterize its pharmacology and structural properties.

We have determined and compared nucleotide excision repair capability of several rat tissues by amethod based on restoration of the transformation activity of UV-irradiated pBlueScript by incubation in repair-competent protein extracts. After 3 hof incubation, plasmidDNAwas isolated and used to transform competent Escherichia colicells. Damaged plasmids showed low transformation efficiency prior to incubation in repair-competent extracts. After incubation the transformation efficiency was restored to different extents permitting calculationof the repair capacity of the extracts....

In several organisms d-cysteine desulfhydrase (d-CDes) activity (EC 4.1.99.4) was measured; this enzyme decomposes d-cysteine into pyruvate, H2S, and NH3. A gene encoding a putative d-CDes protein was identified in Arabidopsis thaliana(L) Heynh. based on high homology to an Escherichia coliprotein called YedO that hasd-CDes activity. The deduced Arabidopsisprotein consists of 401 amino acids and has a molecular mass of 43.9 kDa.

The NMR solution structures of NTX-1 (PDB code 1W6B and BMRB 6288), a long neurotoxin isolated from the venom ofNaja naja oxiana, and the molecular dynamics simulation of these structures are reported. Calculations are based on 1114NOEs, 19 hydrogen bonds, 19 dihedral angle restraints and secondary chemical shifts derived from 1 Hto 13 C HSQC spectrum. Similar to other long neurotoxins, the three-finger like structure shows a double and a triple strandedb-sheet aswell as some flexible regions, particularly at the tip of loop II and the C-terminal tail....

The novel sex-specific potassium channel inhibitor IsTX, a 41-residue peptide, was isolated from the venom of male Opisthacanthus madagascariensis.Two-dimensionalNMR techniques revealed that the structure of IsTX contains a cysteine-stabilizeda/b-fold. IsTX is classified, based on its sequential and structural similarity, in the scorpion short toxin familya-KTx6. Thea-KTx6 family contains a single a-helix and twob-strands connected by four disulfide brid-ges and binds to voltage-gated K + channels and apamin-sensitive Ca 2+ -activated K + channels. ...

Little is known about the functions of DNA polymerasek (Pol k) recently identified in mammals. From the genomic sequence information of rice andArabidopsis, we found that Polkmay be the only member of the X-family in higher plants. We have succeeded in isolating the cDNA and recombinant protein of Polkin a higher plant, rice (Oryza sativaL. cv. Nipponbare) (OsPolk). OsPolkhad activities of DNA polymerase, terminal deoxyribonucleotidyl trans-ferase and deoxyribose phosphate lyase, a marker enzyme for base excision repair. ...

We have isolated a nonfucosylated and three variously fucosylated neutral oligosaccharides from human milk that are based on theiso-lacto-N-octaose core. Their structures were characterized by the combined use of electrospray mass spectrometry (ES-MS) and NMR spectroscopy. The branching pattern and blood group-related Lewis deter-minants, together with partial sequences and linkages of these oligosaccharides, were initially elucidated by high-sensitivity ES-MS/MS analysis, and then their full structure assignment was completed by methylation analysis and 1 H-NMR.
