Pancreatic ductal adenocarcinoma (PDAC) is a ‘difcult-to-treat’ entity. To forecast its prognosis, we introduced a new biomarker, SARIFA (stroma areactive invasion front areas), which are areas at the tumour invasion front lacking desmoplastic stroma reaction upon malignant invasion in the surrounding tissue, leading to direct contact between tumour cells and adipocytes.

A comprehensive understanding of cancer has been furthered with technological improvements and decreasing costs of next-generation sequencing (NGS). However, the complexity of interpreting genomic data is hindering the implementation of high-throughput technologies in the clinical context: increasing evidence on gene–drug interactions complicates the task of assigning clinical significance to genomic variants.

Precision oncology, defined as treatment of patients with targeted therapies matched to specific molecular alterations, has entered routine clinical practice. Particularly in patients with advanced cancer or hemato‑ logic malignancies, for whom no further standard therapies are available, this approach is increasingly applied as last resort option outside of the approved indication.

The lack of effective and accurate predictive indicators remains a major bottleneck for the improvement of the prognosis of patients with hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC). Hepatitis B virus X (HBx) has been widely suggested as a critical pathogenic protein for HBV-driven liver carcinogenesis, while tumor-associated macrophage (TAM) infltration is also closely related to the tumorigenesis and progression of HCC.

Soft-tissue sarcomas (STS) represent a heterogeneous group of rare tumors including more than 70 different histological subtypes. High throughput molecular analysis (next generation sequencing exome [NGS]) is a unique opportunity to identify driver mutations that can change the usual one-size-fits-all treatment paradigm to a patient-driven therapeutic strategy.

Molecular differences in colorectal cancer (CRC) are associated with the metastatic route. Patient survival is mainly driven by metastatic spread thus it is imperative to understand its key drivers to develop biomarkers for risk stratification, follow-up protocols and personalized therapy. Thus, this study aimed to identify genes associated with the metastatic route in CRC.

Exosomes are extracellular vesicles (EVs) derived from endocytic compartments of eukaryotic cells which contain various biomolecules like mRNAs or miRNAs. Exosomes influence the biologic behaviour and progression of malignancies and are promising candidates as non-invasive diagnostic biomarkers or as targets for therapeutic interventions.

Metagenomics holds great promises for deepening our knowledge of key bacterial driven processes, but metagenome assembly remains problematic, typically resulting in representation biases and discarding significant amounts of non-redundant sequence information.

High-throughput gene expression profiles have allowed discovery of potential biomarkers enabling early diagnosis, prognosis and developing individualized treatment. In this study, we present a simple, yet powerful data-driven method which contributes significantly to identification of robust cross-platform gene signature for disease classification of single-patient to facilitate precision medicine.

“Biomarker-driven targeted therapy,” the practice of tailoring patients’ treatment to the expression/ activity levels of disease-specific genes/proteins, remains challenging. For example, while the anti-ERBB2 monoclonal antibody, trastuzumab, was first developed using well-characterized, diverse in vitro breast cancer models (and is now a standard adjuvant therapy for ERBB2-positive breast cancer patients), trastuzumab approval for ERBB2-positive gastric cancer was largely based on preclinical studies of a single cell line, NCI-N87.

NCI’s Division of Cancer Prevention set out 7 years ago to create a strong, investigator-driven network to conduct translational research to identify tests for early cancer and cancer risk. In 2000, the Early Detec- tion Research Network (EDRN) became a fully funded group of 28 grantees focused on the overarching goal of creating validated biomarkers ready for large-scale clinical testing. Today, EDRN is a nationwide, interdisciplinary group of established partnerships among scores of institutions and hundreds of individuals working to advance the science for public benefit. ...

EDRN is at the forefront of technology-driven research on the use of biomarkers for the early detection of cancer. By identifying and validat- ing biomarkers, such as novel proteins or changes in gene expression, it is possible to measure an individual’s disease risk, progression of disease, or response to therapy. Ultimately, EDRN research will aid in prevention and in early therapeutic intervention, based on early detection of disease.

While there has been an increase in development assistance for health in general, this aid is volatile — often driven by profits and politics. Donor governments are susceptible to the political agendas of their parliaments, plac- ing a focus on short term results. Sustained health development does not fit this profile. Part of the solution lies in the harmonization or pooling of funds, focused around a single country plan, that is flexible and includes funding for both recurrent and capital expenditures.

The UK is an open economy and an active and successful international trader and investor. But the UK’s reliance on debt-financed consumer and government spending, and on the financial sector, has driven growing imbalances in the UK economy. According to the OECD, by 2007 the UK had the largest structural budget deficit in the G7 group of countries. The current account deficit was around 2¾% of GDP in 2007, a figure that was offset by a 2¼% surplus on trade in financial services.

Historically, most cancer research has been conducted through small independent projects initiated by individual investigators with relatively small research groups. Such research is driven by focused hypotheses addressing specific biological questions. There will always be a need for this traditional approach to research; in recent years, however, it has also become more feasible to undertake projects on a broader and larger scale, thereby developing extensive pools of data and research tools that can facilitate those more conventional efforts....
