Ebook "The plant cell wall: Methods and protocols" describes some of the methods which are currently applied to investigate the many aspects of the plant cell wall including its structure, biochemical composition, and metabolism, to name but a few. Each chapter is written by leading experts in cell wall research and is written with the aim that the protocol(s) can be carried out by someone without previous experience in that particular method or specifically in cell wall research.

Ebook "Green biomass pretreatment for biofuels production" emphasises the tactical mobile and on-farm scales applications that use green pretreatments and processing technologies without the need of on-site waste treatment. Because of the varieties of different biomasses, no single pretreatment is expected to be the universal choice. Some of the pretreatment methods present niche applications are also discussed.

A bifunctional glycoside hydrolase GH78 from the ascomycete Xylaria polymorpha (XpoGH78) with catalytic versatility to both glycosides and esters, may be beneficial for the efficient degradation of plant cell-wall complexes containing both diverse sugar residues and esterified structures to apply in, for example, the production of biofuels from plant feedstock.

In this paper, we show a detailed look at the barley XTH gene family, including detailed bioinformatics analyses of conserved protein motifs and structure, phylogenetic relationships, and a comparison of the expression patterns during germination and seedling growth using RT-qPCR and RNA-Seq analyses. Overall, there was a good correlation between RT-qPCR and RNA-Seq data for similar tissues. RNA-Seq data showed many different expression profile patterns, from broad, high level expression for some XTH genes to highly tissue specific patterns for others.

To understand the architecture of the plant cell wall, it is of importance to understand both structural characteristics of cell wall polysaccharides and interactions between these polysaccharides. Interactions between polysaccharides were studied in the residue after water and chelating agent extraction by sequential extractions with H2O and alkali.

In wheat endosperm, mannan, is poorly documented. Nevertheless, this hemicellulosic polysaccharide might have a determinant role in wheat grain development since, in Arabidopsis thaliana, mutants with a reduced amount of mannan show an altered seed development.

Feruloyl esterase catalyze the hydrolysis of a wide variety of glycosidic linkages between two or more carbohydrates or between a carbohydrate and a non-carbohydrate moiety. Thereby, they play a role in the initial degradation of lignocelluloses, which may be advantageous for the efficient degradation of the plant cell-wall complex that contains both diverse sugar residues and esterified structures.

In the present study, wrinkled surfaces were discovered on the cell wall thickenings of helical vessels in the lilac style for the first time. The structure of the wrinkles was formed by cellulose microfibrils, with convex ridges composed of crystalline cellulose and concave grooves composed of amorphous cellulose. The cellulose microfibrils were oriented with the helical cell wall thickenings. The wrinkle morphology was due to the aligned crystalline region of the microfibrils.

The results of the physiological parameters showed that their profile changes were consistent with those of the proteome analysis. The results of the proteome and physiological parameters showed that K. candel roots could resist highsalinity stress by maintaining a normal Embden-Meyerhof-Parnas and tricarboxylic acid (EMP-TCA) pathway, increasing the activities of various antioxidant enzymes and antioxidant contents, stabilizing the cell wall structure, and accumulating secondary metabolites such as triterpenoids.

Fragaria vesca or ‘woodland strawberry’ has emerged as an attractive model for the study of ripening of non-climacteric fruit. It has several advantages, such as its small genome and its diploidy. The recent availability of the complete sequence of its genome opens the possibility for further analysis and its use as a reference species. Fruit softening is a physiological event and involves many biochemical changes that take place at the final stages of fruit development; among them, the remodeling of cell walls by the action of a set of enzymes.

Xyloglucan endotransglycosylase/hydrolases (XTH) can disrupt and reconnect the xyloglucan chains, modify the cellulose-xyloglucan complex structure in the cell wall to reconstruct the cell wall. Previous studies have reported that XTH plays a key role in the aluminum (Al) tolerance of tea plants (Camellia sinensis), which is a typical plant that accumulates Al and fluoride (F), but its role in F resistance has not been reported.

Pectin is a major component and structural polysaccharide of the primary cell walls and middle lamella of higher plants. Pectate lyase (PEL, EC 4.2.2.2), a cell wall modification enzyme, degrades de-esterified pectin for cell wall loosening, remodeling and rearrangement. Nevertheless, there have been few studies on PEL genes and no comprehensive analysis of the PEL gene family in cotton.
