Characterization of pectin

The degree of esterification and methoxyl contents were varying depending upon the maturity stages. Therefore, the premature lemon pomace can be considered as rich source of pectin in terms of yield, methoxyl content, degree of esterification and anhydrouronic acid content.

New pectin derivative (Pec-MA) was obtained in specific reaction conditions. The presence of maleoyl groups in Pec-MA structure was confirmed by 1H NMR and FTIR spectroscopy. The substitution degree of Pec-MA (DS = 24%) was determined by 1H NMR.

About 0.5 ton of coffee pulp is generated for each ton of coffee cherry processed. In the present study, this waste was investigated as a source of pectin. Coffea arabica L. pulp was dried, treated with ethanol and the pectin extracted with 0.1 M HNO3 (14.6 % yield). Chromatographic, colorimetric and spectroscopic methods were used for pectin characterization.

In this work we have efficiently extracted and characterized pectin from different tissues of astringent (AS) and non-astringent (NAS) persimmon fruits (peel, pulp, whole fruit) for the first time. The highest pectin extraction (≥7.2%) was carried out at 80 ◦C, 120 min with 1.5% sodium citrate in peel of both AS and NAS persimmon samples.

Comprehensive chemical characterization of nine mono-varietal apple pomaces obtained from the production of ciders with PDO is described. They were rich in essential minerals, fibers (35–52.9 %), and polyphenols. High levels in GalA (11.8–21.6 %), revealed the suitability of these apple pomaces as efficient sources of pectins.

This study primarily investigated the possibility of extracting pectin from orange peels using water as a green solvent. The pectin powder was characterized. The results show that the extracted pectin had acceptable moisture content (8,21%) and high purity with the total anhydrouronic acid content of 82,7%.

Pectin is a heterogeneous complex polysaccharide found in the primary cell wall of most cells. In this work, lowmethoxyl pectin was extracted from pumpkin (Cucurbita moschata). The obtained pectin was characterized by GPC FTIR and NMR spectra. The obtain results showed that structural feature of the pectin is a main chain of poly-α(1→4)- D-galacturonic acid and occasionally interrupted by α(1→2)-linked α-L-rhamnopyranose residues and branches Galp is linked to GalpA at C-2 position.

Fruit wastes such as citrus peel, mango peel, apple pomace and banana peel were collected from fruit juice manufacturing industries and were subjected to pectin extraction. The results revealed that citrus peel is containing higher pectin content of about 24.5%. Characterization of different fruit waste were also done.

Pectin methylesterase (PME) is a hydrolytic enzyme that catalyzes the demethylesterification of homogalacturonans and controls pectin reconstruction, being essential in regulation of cell wall modification. During fruit ripening stage, PME-mediated cell wall remodeling is an important process to determine fruit firmness and softening.

Banana pseudostem comprises several polymers such as cellulose, hemicellulose, pectin and lignin that constitute fibers with good mechanical properties. These sugars can be used for production of various organic acids and alcohol. With the availability of such huge biomass as substrate, a wide range of microorganisms like bacteria and fungi grow on it. Lactic acid bacteria can grow on such sugars and can be isolated from banana pseudostem. In present study, lactic acid bacteria (LAB) were isolated from banana pseudostem core using MRS agar.

TheAspergillus nigerb-1,4-endogalactanase encoding gene (galA) was cloned and characterized. The expression ofgalA in A. nigerwas only detected in the presence of sugar beet pectin,D-galacturonic acid andL-arabinose, suggesting that galAis coregulatedwithboth thepectinolyticgenes aswell as the arabinanolytic genes. The corresponding enzyme, endogalactanase A (GALA), contains both active site resi-dues identified previously for thePseudomonas fluorescens b-1,4-endogalactanase.

An intracellular pectinolytic enzyme, PelB (TM0437), from the hyperther-mophilic bacteriumThermotoga maritimawas functionally produced in Escherichia coliand purified to homogeneity. PelB belongs to family 28 of the glycoside hydrolases, consisting of pectin-hydrolysing enzymes. As one of the few bacterial exopolygalacturonases, it is able to remove monogalac-turonate units from the nonreducing end of polygalacturonate.
