Results of the batch fermentation showed an earlier production of CHO enzyme with 4% increase in activity compared to that obtained from shake flask cultures indicated that Streptomyces sp. strain NHIA_CH5 could be a potential source of CHO enzyme for commercial purpose.

A colorimetric chemosensor for hydrogen peroxide (H2O2) and biosensor for cholesterol is developed in this work following a ‘label-free and reagentless’ concept. In this strategy, silver nanoparticles (AgNPs) are used as a bifunctional probe, namely, a chemical capture probe for H2O2 recognition and a signal probe for displaying detection results. Cholesterol oxidase is a bacterial FAD-containing flavooxidase that catalyzes the first reaction in cholesterol catabolism. Indeed, this enzyme catalyzes two reactions: the oxidation of the C3 -OH group of cholesterol (and other sterols) to give cholest-5-en-3-one; and its isomerization to cholest-4-en-3-one. In the past several years, the structural and functional characterization of choles-terol oxidase has been developed together with its application as a biological tool.

An important aspect of catalysis performed by cholesterol oxidase (3b-hy-droxysteroid oxidase) concerns the nature of its association with the lipid bilayer that contains the sterol substrate. Efficient catalytic turnover is affected by the association of the protein with the membrane as well as the solubility of the substrate in the lipid bilayer.

Cholesterol oxidases are bifunctional flavoenzymes that catalyze the oxida-tion of steroid substrates which have a hydroxyl group at the 3bposition of the steroid ring system. The enzyme is found, in a wide range of bacte-rial species, in two forms: one with the FAD cofactor bound noncovalently to the enzyme; and one with the cofactor linked covalently to the protein.
