A1AT inhibits the activity of many proteases, but its main task is to protect the lungs from the attack of neutrophil elastase. In an autosomal hereditary disease known as alpha-1 antitrypsin deficiency, the A1AT level in blood serum decreases, increasing the risk of developing emphysema, liver apoptosis, and liver cancer.   • The stabilization and three-dimensional modeling of mutant proteins were also determined by using the I-Mutant 2.0 and Project HOPE webservers, respectively. In total, 772 missense SNPs were found in the APAF1 gene from the NCBI dbSNP database, 18 SNPs of which were demonstrated to be deleterious or damaging. Of those, 13 SNPs had a decreasing effect on protein stability, while the other 5 SNPs had an increasing effect. Based on the modeling results, some dissimilarities of mutant type amino acids from wild-type amino acids such as size, charge, and hydrophobicity were revealed.

  • High performance liquid chromatography was applied for the determination of tryptophan in food. The sample was hydrolyzed in a duration from 16 to 24 hour by protease enzyme at 50o C. The extract was separated on the C8 reversed phase column with mobile phase of NaH2PO4 50mM pH 2.3: Methanol (82:18; v/v). The linearity of the method was kept in the range of 0.5 - 50 mg/L.   • Response surface methodology (RSM) was used to determine optimal condition for continuous overflow proteolytic hydrolysis of spent brewer’s yeast. The optimal conditions for obtaining high degree of hydrolysis and low bitterness are determined as followings: ratio of enzyme mixture (alcalase 7.5 U/g and flavourzyme 10 U/g), pH at 7.5, hydrolysis temperature at 51o C and hydrolysis time of 9 hours.
