The miR-144-3p was discovered to upregulate in osteoarthritis. However, the function of this molecule is still poorly understood. Identification of microRNA target genes could shed a light on miRNA functions. In this project, we aim to identify a direct target of miR-144-3p. Potential targets of miR-144-3p were predicted by bioinformatic algorithms.

The effect of CoA on the characteristic light decay of the firefly luciferase catalysed bioluminescence reaction was studied. At least part of the light decay is due to the luciferase catalysed formation of dehydroluciferyl-adenylate (L-AMP), a by-product that results from oxidation of luciferyl-adenylate (LH2-AMP), and is a powerful inhibitor of the bioluminescence reaction (IC50¼6nm).

We introduce a new application of firefly luciferase (EC 1.13.12.7). The firefly luciferases belong to a large superfamily that includes rat liver long-chain acyl-CoA synthetase (LACS1). LACS1 is the enzyme that is involved in the deracemization process of 2-arylpropanoic acid and catalyzes the enantioselective thioester formation of R-acids.

The activating and stabilizing effects of inorganic pyrophosphate, tripoly-phosphate and nucleoside triphosphates on firefly luciferase biolumines-cence were studied. The results obtained show that those effects are a consequence of the luciferase-catalyzed splitting of dehydroluciferyl-adenyl-ate, a powerful inhibitor formed as a side product in the course of the bio-luminescence reaction.
