Part 1 book "PCR primer design" includes content: Fast masking of repeated primer binding sites in eukaryotic genomes, design of primers and probes for quantitative real time PCR methods, low concentration initiator primers improve the amplification of gene targets with high sequence variability, degenerate primer design for highly variable genomes,....and other contents.

Compared to RNA-sequencing transcript differential analysis, gene-level differential expression analysis is more robust and experimentally actionable. However, the use of gene counts for statistical analysis can mask transcript-level dynamics. We demonstrate that ‘analysis first, aggregation second,’ where the p values derived from transcript analysis are aggregated to obtain gene-level results, increase sensitivity and accuracy.

Cell-to-cell variation and heterogeneity are fundamental and intrinsic characteristics of stem cell populations, but these differences are masked when bulk cells are used for omic analysis. Single-cell sequencing technologies serve as powerful tools to dissect cellular heterogeneity comprehensively and to identify distinct phenotypic cell types, even within a ‘homogeneous’ stem cell population.

Microarray technology, as well as other functional genomics experiments, allow simultaneous measurements of thousands of genes within each sample. Both the prediction accuracy and interpretability of a classifier could be enhanced by performing the classification based only on selected discriminative genes.

A food may test positive for glucose but not have enough glucose to taste sweet, or the sweet flavor may be masked, e.g. by a sour flavor. Sweet foods may have little glucose, but significant amounts of other sugars (or artificial sweeteners that bind to taste receptors for sugars). When there is a small amount of a type of molecule (e.g. fat in pretzels) the tests used in this activity may not be sensitive enough to read positive. The results of the protein test for pretzels may be somewhat ambiguous, but if a little bit of pretzel is crushed...
