Ebook "The hypothalamic-pituitary-adrenal axis in health and disease: Cushing’s syndrome and beyond" reviews the role of cortisol in the human body, focusing on the effects of excess cortisol due to Cushing’s syndrome as well as the role of the HPA axis in metabolism, inflammation, and neuropsychiatric function. The volume will cover basic mechanistic data, clinical outcomes data, and novel therapies.

Sarcoidosis is a multisystemic granulomatous disease of unknown origin. It is characterized by abnormal activation of lymphocytes and macrophages with the formation of granulomas. Most cases have asymptomatic pulmonary involvement. In case of symptoms, they have an excellent response to glucocorticoid therapy.

Adrenal insufficiency is a life-threatening condition, and advanced gastric cancer is associated with very poor prognosis. Adrenalectomy and/or metastatic invasion of the adrenal glands can cause primary adrenal insufficiency, which in turn can present with symptoms mimicking advanced cancer.

Combination therapy with glucocorticoids and adjunctive immunomodulating drugs has been generally accepted as a standard treatment regimen for meningoencephalomyelitis of unknown etiology (MUE). We hypothesized that treatment with MMF as an adjunctive agent along with glucocorticoids would be effective and well-tolerated protocol in dogs with MUE.

There is evidence that postmenopausal women with rheumatoid arthritis (RA) on glucocorticoid (GC) therapy and bisphosphonate (BP) have an increased risk for atypical subtrochanteric and atypical diaphyseal femoral fracture (AFF). The underlying mechanism has not been elucidated so far.

Due to broad range of effects, corticosteroids are frequently used in veterinary practice. Wide ranges of biological effects are mediated via different dose and potency of different glucocorticoids. Intensive short-term glucocorticoid use can decrease the morbidity and mortality of potentially fatal conditions but in long run their use can lead to immunosuppression, duodenal ulceration, pancreatitis, adrenal atrophy, hyperadrenocorticism, glucose intolerance and many more diseases.

Glucocorticoid therapy, especially at higher doses, is associated with significant adverse side effects including osteoporosis. Leptin, secreted from adipose tissue, has diverse effects on bone tissue regulation. As glucocorticoids stimulate leptin synthesis and secretion directly in adipose tissue we hypothesised that dexamethasone (DEX) induced osteoporosis may, in part, be mediated by an osteoblast dependent leptin-leptin receptor pathway.

Modification of the surface of these vectors is a key element for their successful research and clinical use. Chapter three discusses the methods to modify surfaces of retroviral vectors, and the applications for surface modification of retroviral vectors, such as targeting and immune modulation. Chapter four reviews the role of the nuclear glucocorticoid receptor in controlling retroviral infection and function, and highlights its potential importance in retroviral-based gene therapy applications....
