Green gram is a widely cultivated pulse crop rich in protein, essential amino acids and vitamin-B. The quality and quantity of DNA are very important for amplification by PCR. Although quantity of DNA required per reaction in PCR is very low, quality is very crucial. Also, to carry out large number of PCR reactions for genotyping, a good amount of DNA is required. Presence of contaminants like phenols makes it difficult to get good quality DNA from mungbean.

The green gram (Vigna radiata L. Wilczek) is one of the important pulse crop because of its adaptation to short growth duration, low water requirement, soil fertility and is favoured for consumption due to its easy digestibility and low production of flatulence (Shil and Bandopadhya, 2007).

Molecular characterization is helpful in understanding the phylogenetic relationship among various germplasm to reveal the genetic diversity among the used parental genotypes. Among several efficient methods for revealing genetic variability within and among plant populations, one of the most widely applied methods is ISSR marker analysis. ISSR, markers are commonly used because they are quick, simple and environment non-sensitive enabling genetic diversity analysis in several types of plant material like natural populations, population in breeding programmes.

An experiment was conducted during summer season of 2017 at the college farm, Navsari Agricultural University, Navsari to study the “Production potential of green gram [Vigna radiata (L.) Wilczek] varieties with different nutrient management under south Gujarat condition”. The soil of the experimental field was clayey in texture having medium to poor drainage, low in available nitrogen and organic carbon, medium in available phosphorus, potassium and sulphur.

The present investigation entitled “Principal component analysis in rainfed green gram genotypes [Vigna radiata (L.) Wilczek]” was carried out to determine the relationship and genetic diversity among 16 green gram genotypes using principal component analysis for various characters during Kharif, 2019 at Agricultural Research Station, Fatehpur - Shekhawati, Sikar (Rajasthan) under rainfed conduction. Principal component analysis (PCA) depicted that three components (PC1 to PC3) accounted for about more than 90% of the total variation for different characters.

The present experiment comprised of three parents Meha, GJM-1006 and GJM-1008 their 30 F4 progenies derived from the crosses Meha x GJM-1006 (17 progenies) and Meha x GJM-1008 (13 progenies). Seed yield per plant was strongly and positively associated with the yield components like days to 50 % flowering, plant height, primary branches per plant, days to maturity, pods per plant, 100 seed weight, seeds per plant and clusters per plant in the two segregating populations except days to 50 % flowering and days to maturity in the cross Meha x GJM-1006.

In the present investigation, a simple, rapid and efficient in vitro regeneration system was developed in green gram (Vigna radiata L. Wilczek) cv. IPM-02-03 using cotyledonary explants excised from a 4 day old seedling. Organogenic calli were developed from cotyledonary tissues within 4-6 weeks of culture, supplemented with 2.5 mg/L BAP and 1.5 mg/L NAA. Shoot regeneration was achieved from shoot tips and cotyledonary node on MS medium was supplemented with BAP 2.0 mg/l

Field screening of 50 green gram genotypes was done at Economic Botany (EB)-II, OUAT, Bhubaneswar, Odisha during summer 2015 to identify the Mungbean Yellow Mosaic Virus (MYMV) resistant genotypes. The disease screening for MYMV was carried out under natural field condition using infector rows of highly susceptible genotype, KPS 2, in the hot spot area during summer 2015 when vector population was high.

Ten genotypes as lines and three genotypes as testers of mungbean were investigated for combining ability analysis through crosses developed into line x tester passion. The study revealed that the gene action involved in controlling traits viz., plant height, number of pods per plant, number of seeds per pod and grain yield per plant was non-additive; while the characters viz., number of branches per plant and 100-grain weight had additive gene action. Genotype KM 2264 was found good general combines for most of the yield attributing traits.

Fifty-six germplasm lines of green gram [Vigna radiata (L.) Wilczek] were used to determine the extent of genetic diversity using Mahalanobis D2 statistic. The analysis was carried out for 20 quantitative traits. The D2 statistic performed on 20 quantitative traits grouped all 56 genotypes into eight divergent clusters. Cluster V (8559.766) had maximum intra-cluster distance while inter-cluster distance was highest between clusters III and VI(329997.

Mungbean [Vigna radiata (L.) Wilczek] known as green gram is one of the major fastgrowing, warm-season crop that is primarily cultivated in developing countries of Asia. The production of mungbean is severely constrained by mungbean yellow mosaic virus (MYMV) caused by begomo viruses, which is transmitted by whitefly, Bemesia tabaci. The absence of resistant/tolerant sources against MYMV disease leads to tremendous crop yield losses.
