  • The highest significant values of total soluble sugars and their fractions were noted in explant stage. Embryogenic callus stage induced a significant increase in total soluble sugars, free amino acids, phenols and indoles concentrations as compared to callus and mature somatic embryo.

  • Phalaenopsis is one of the most remarkable commercial genera in Orchidaceae family. Using immature explants for tissue culture of orchid results in undesired characteristics such as genetic variation and long-lasting juvenility. In this study, we optimized a protocol for somatic embryogenesis of Phalaenopsis amabilis cv. Jinan, by using mature thin cell layer (TCL) explants to minimize immature explant limitations. We examined somatic embryogenesis from two types of mature explants, viz.

  • Barley is known to be recalcitrant to tissue culture, which hinders genetic transformation and its biotechnological application. To date, the ideal explant for transformation remains limited to immature embryos; the mechanism underlying embryonic callus formation is elusive.

  • Somatic embryogenesis based plant regeneration is a pre-requisite for genetic transformation of banana. Mass multiplication of banana through somatic embryogenesis is also viewed as an economically viable alternative to commonly used shoot tip based method. Banana somatic embryogenesis response was studied in three popular banana cultivars, Grand Naine, Rasthali and Hill banana „Virupakshi‟. Immature male flower bud explant was used for embryogenic callus induction.

  • The CRISPR/Cas9 system has been used predominantly for precise editing of the plant genomes. In the present study, we have made an application of CRISPR/Cas9 system for targeted mutagenesis in rice(Oryza sativa L.) targeting the Phytochrome and Flowering Time 1 (PFT1) gene known to be involved in disease susceptibility in Arabidopsis for rootinfecting hemibiotrophic fungal pathogen Fusarium oxysporum. Hence, to confirm the role of OsPFT1 gene in rice sheath blight disease, CRISPR-Cas9 gene editing tool is being employed for generating OsPFT1 gene knock out in rice.

  • A protocol for in vitro propagation of Muscari mirum Speta, an endangered geophyte of the family Hyacinthaceae, was developed. Bulb scale, scape, leaf, and immature embryo explants were cultured on different nutrient media compositions supplemented with various concentrations of plant growth regulators (BAP, NAA, TDZ, Picloram, 2,4-D). Scape explants produced callus only.

  • In this study, to evaluate the ability to accept transgenes of the two cassava cultivars KM94 and KM140, which are grown widely in Vietnam, A. tumefaciens bacterial strains C58/pGV2260, EHA105 and LBA4404 containing vector pCB-gusplus or pPIPRA558 haboring selectable marker gene gus/gusplus, were co-inoculated with explants from four selected sources, including (1) immature leaves, (2) shoot apexes, (3) callus, and (4) somatic embryo cotyledons.
