  • A study was carried out to establish efficient and reliable in vitro propagation protocol for Gisela 5 (Prunus cerasus x Prunus canescens) - clonal cherry rootstock. Axillary and terminal buds of ‘Gisela 5’ were collected in the spring and cultured on MS medium. After the in vitro cultures were sufficiently established, experiments were conducted to assess the influence of different levels of benzyladenine (BA), kinetin (Kin), thidiazuron (TDZ), gibrellic acid (GA3) and indole-3-butyric acid (IBA) on in vitro shoot multiplication.

  • The present study was conducted to assess the effect of composite culture of Azotobacter and Phosphate solubulizing bacteria with different combinations of organic, inorganic fertilizations on growth performance of in vitro propagated Banana, cv. Grand Naine. The in vitro rooted plantlets were hardened and acclimatized by using different treatments (five treatments comprising of inoculation of composite culture, vermicompost and Inorganic fertilization) carried out in CRD replicated four times.

  • Plant tissue culture is the method of culturing the plant cells or tissues under controlled aseptic conditions on a solid or liquid medium. It has value in basic research for the production of true to type, disease free and micro-propagated plants. Besides this also has commercial application. This review work outlines the work done on pertaining to various aspects of in vitro micro-propagation, regeneration, rooting, hardening, in potato.

  • In vitro regeneration and in planta transformation protocols were standardized for the black gram (Vigna mungo L.) cv. VBN 3, an important grain legume. An efficient regeneration protocol was developed using 1-week-old de-embryonated cotyledon as explants. The maximum frequency of shoot regeneration (72.6%), multiple shoot induction (6.0 ± 0.3) and shoot elongation was achieved on MS medium supplemented with 3.0 mg/l 6-benzyl amino purine (BAP) after 4 weeks of culture. Up to 65% of the regenerated shoots were rooted on MS medium containing 0.

  • Coconut has a wide range of applications from culinary to industrial areas. Loss of coconut trees due to the devastating effect of deadly diseases and the natural aging of the plants demand rejuvenation of orchards with elite seedlings. A conventional breeding program is not sufficient to meet the growing need for healthy planting materials. However, in vitro culture studies can pave the way for mass multiplication. In this study, the effect of TDZ on sliced mature embryo explants through direct organogenesis of coconut var. Chowghat Orange Dwarf (COD) was investigated.

  • Conventional breeding involves time taking procedure for pyramiding of genes and screening of crops for the same. Double haploid (DH) production can reduce the time of production of resistant crops in less time period. Twenty one plants that survived after the hardening procedure were screened phenotypically and molecularly for BB resistant genes.

  • A study was carried out to establish efficient hardening strategy for in vitro raised plantlets of four grape genotypes. In vitro grown plantlets of Pusa Navrang, Hybrid 76-1 (Hur x Cardinal), Pearl of Csaba and Julesky Muscat were hardened in vitro using hardening medium consisting coco peat + vermiculite + perlite (2:1:1) ratio and two different types of hardening pots viz. glass jars with polypropylene cap (GJPP) and plastic pots with polyethylene cover (PPPC).

  • The present investigation entitled as “Effect of hardening media and different cross combinations on physiological parameters of in vitro cultured citrus plantlets” was carried out in the Division of Fruits and Horticultural Technology, ICAR-IARI, New Delhi. The experiment consisted of embryo rescued plantlets from cross pollination (control pollination) and open pollination (uncontrolled pollination) and two hardening media HM1(Coco peat: Perlite: Vermiculite, 1:1:1) and HM2 (Coco peat: Perlite: Vermiculite, 2:1:1).

  • In this study, the effect of Vesicular Arbrbusular Mycorrhizae (VAM) on the ex vitro hardening of marigold was investigated. A protocol was generated for ex vitro hardening of in vitro raised plantlets using VAM consortia (Glomus intraradices, Glomus mosseae and Scutellospora spp.) in different media mixtures, and their effect was observed on plant survival and growth. Highest percent survival (94.00), maximum plant height (39.00 cm), maximum plant spread (30.50 cm), maximum stem diameter (0.97 cm), maximum shoot fresh weight (0.75 g), maximum root fresh weight (0.

  • The highest shoot length (13.1 cm) with 100% frequency was achieved on medium containing 1.0 mg/L GA3. In vitro proliferated shoots were then excised from the shoot clumps and transferred to the rooting medium containing different concentrations of indole butyric acid (IBA; 0.5-2.0 mg/L) and indole acetic acid (IAA; 0.5-2.0 mg/L) alone. Among these, the highest root proliferation was obtained in the medium containing 1.5 mg/L IBA.

  • An efficient and reproducible plant regeneration system was developed from cells or tissues of agronomically important Indian sorghum genotypes including 2 commercial cultivars (NSH27 & K8) of Sorghum bicolor (L.) Moench. Callus induction and plant regeneration were achieved on transverse thin cell layers (tTCL) of roots from aseptically germinated 7-day-old seedlings.
