In vitro shoot multiplication rate

A study was carried out to establish efficient and reliable in vitro propagation protocol for Gisela 5 (Prunus cerasus x Prunus canescens) - clonal cherry rootstock. Axillary and terminal buds of ‘Gisela 5’ were collected in the spring and cultured on MS medium. After the in vitro cultures were sufficiently established, experiments were conducted to assess the influence of different levels of benzyladenine (BA), kinetin (Kin), thidiazuron (TDZ), gibrellic acid (GA3) and indole-3-butyric acid (IBA) on in vitro shoot multiplication.

In addition, the effects of explant age and lighting condition on shoot regeneration were also investigated. After 8 weeks of culture, the results showed that shoot regeneration rate (100%) and shoot multiplication coefficient (13.33) of the in vitro leaf-tTCL-4 were higher than those of other treatments and control. The shoot regeneration rate of P. edulis Sims. also varied with the change of explant age. The highest shoot regeneration rate (100%) was obtained from leaf explants of 1.5-month-old shoots after 8 weeks of culture.

Suitable medium for rooting was MS + 0.5 mg/L IBA with root shoots at 97.33%, average roots at 5.29 roots/shoot after 4 weeks. On organic substrat, 70% coconut fiber and 30% composted cow manure gave the highest survival rate of 91.33%. The plants grew and developed well during the nursery stage.

The cultivar Robusta forms the mainstay of commercial banana cultivation in India. An efficient micropropagation system with high multiplication rate will boost banana cultivation and assist banana based industries in the country. The shoot apices of banana cv. Robusta were cultured on two basal media MS and B5 supplemented with different concentrations and combinations of plant growth regulators (PGR). The tissue culture responses were observed for shoot multiplication and effect of subculture on rate of shoot multiplication till 8th subculture.

Banana (Musa spp.) cv. Rajapuri Bale (AAB) is a popular cultivar of banana grown in Northern parts of Karnataka especially in Bagalkot, Bijapur and Belgaum. The demand for the planting material of this cultivar is high and tissue cultured plantlets is not available because of the inherent problem in the initial establishment of the culture and multiplication rate is also low.

In vitro regeneration and in planta transformation protocols were standardized for the black gram (Vigna mungo L.) cv. VBN 3, an important grain legume. An efficient regeneration protocol was developed using 1-week-old de-embryonated cotyledon as explants. The maximum frequency of shoot regeneration (72.6%), multiple shoot induction (6.0 ± 0.3) and shoot elongation was achieved on MS medium supplemented with 3.0 mg/l 6-benzyl amino purine (BAP) after 4 weeks of culture. Up to 65% of the regenerated shoots were rooted on MS medium containing 0.

Marigold is native to Mexico and one of the commercial loose flower crops in India. In general it is commonly propagated through seeds, but some ornamentally high valued petaloid and gynomonoecious lines can only be maintained through vegetative propagation. Initial in vitro axenic culture establishment, poor multiplication rates, excess callusing and vitrified cultures are the major hindrances in its commercial micro-propagation.

Gladiolus (Gladiolus hybrida) is an important ornamental plant cultivated world over for its attractive spikes. The commercial cultivation of Gladiolus is based on natural multiplication of corms and cormels. In vitro propagation techniques, is superior to conventional propagation and produces disease free plants in huge quantities. In vitro regeneration of gladiolus cultivars, Sylvia, White Prosperity and Amsterdam was achieved using shoot bud of cormels as an explant. Sylvia variety recorded the best for callus induction and regeneration than other two varieties.

Micropropagation of Indian Blackberry (Syzygium cumini L.) species locally called as Jamun, has always been a challenging task. In India, Jamun tree is an underutilized minor fruit crop species possessing a high medicinal and nutritional value. An efficient protocol for rapid shoot proliferation of Jamun (cv. Rajamun) from nodal segment of locally grown mature trees has been developed. Experiments were conducted at Biotech Centre, Dr. PDKV, Akola during 2016-18. The Lloyd and McCown woody plant medium (WPM) used as a basal media supplemented with growth regulators at varying concentrations.

A method for micropropagation was developed for Dendrocalamus asper, an economically and environmentally important bamboo. Disinfected seeds were cultured in flasks containing 20 ml of Murashige and Skoog’s medium (MS) supplemented with BA (1.0-7.0 mg l-1 ) or kinetin (1.0-7.0 mg l-1 ). Multiple shoots (6.53) were formed on MS medium supplemented with 3.0 mg l-1 BA and reached 1.49 cm in length. Continuous shoot proliferation was achieved on a MS medium supplemented with BA (1.0-7.0 mg l -1 ). The multiplication rate of 3.30 fold was achieved on MS medium supplemented with 3.0 mg l-1 BA.
