Acute myocardial infarction may occur following cocaine abuse. Cocaine-induced infarction is particularly common in younger patients aged 18 to 45 years old. Patients may or may not have evidence of coronary artery disease at the time of symptom presentation.

The global rise in aging population led to the increase of the number of elderly patients with lung cancer. Due to the impairment of the function of multiple organs, the work-up and treatment NSCLC for older patients become more difficult than adults.

Amyloid protein (Ab1–40) aggregation and conformation was examined using native and sodium dodecyl sulfate/ polyacrylamide gel electrophoresis, and the results com-pared with those obtained by atomic force microscopy, and with Congo red binding, sedimentation and turbidity assays. The amount of Ab aggregation measured was different, depending upon the method used. Incubation for 15 min at pH 5.0 or in the presence of Fe 2+ ,Cu 2+ or Zn 2+ did not alter the level of Aboligomers observed on SDS and native gels....

Ascorbic acid is often regarded as an antioxidantin vivo, where it protects against cancer by scavenging DNA-dam-aging reactive oxygen species. However, the detailed mech-anism of the action of ascorbic acid on genetic DNA is still unclear. We examined the effect of ascorbic acid on the higher-order structure of DNA through real-time observa-tion by fluorescence microscopy. We found that ascorbic acid generates a pearling structure in single giant DNA molecules, with elongated and compact regions coexisting along a molecular chain. ...

It has been well documented that both endogenous inflammatory mediator advanced glycation end products (AGEs) and exogenous inflammatory inducer lipopolysaccharide play key roles in the initiation and development of inflammatory diseases. However, the combined inflammation-stimulatory effect of AGEs and lipopolysaccharide on endothelial cells, and, further-more, the underlying signal transduction pathways involved, have not been fully elucidated.

he tumor suppressor, breast cancer susceptibility gene 1 (BRCA1), plays an integral role in the maintenance of genome stability and, in particular, the cellular response to DNA damage. Here, the emerging role of BRCA1 in nonhomologous end-joining-mediated DNA repair following DNA dam-age will be reviewed, as well as the activation of apoptotic pathways.

Oxidative damage to mtDNA is associated with excessive reactive oxygen species production. The mitochondrial common deletion (mtDNA 4977-bp and 4834-bp deletion in humans and rats, respectively) is the most typical and frequent form of mtDNA damage associated with aging and degenera-tive diseases.

Huntington’s disease (HD) is an inherited neurodegenerative disorder characterized by cortico-striatal dysfunction and loss of glutamate uptake. At 7 weeks of age, R6/2 mice, which model an aggressive form of juvenile HD, show a glutamate-uptake deficit in striatum that can be reversed by treatment with ceftriaxone, a b-lactam antibiotic that increases GLT1 expression. Only at advanced ages ( 11 weeks), however, do R6/2 mice show an actual loss of striatal GLT1. Here, we tested whether ceftriaxone can reverse the decline in GLT1 expression that occurs in older R6/2s.

Acute Myeloid Leukemia: Treatment Treatment of the newly diagnosed patient with AML is usually divided into two phases, induction and postremission management (Fig. 104-2). The initial goal is to quickly induce CR. Once CR is obtained, further therapy must be used to prolong survival and achieve cure. The initial induction treatment and subsequent postremission therapy are often chosen based on the patient's age.

Vitamin E Deficiency: Treatment Symptomatic vitamin E deficiency should be treated with 800–1200 mg of αtocopherol per day. Patients with abetalipoproteinemia may need as much as 5000–7000 mg/d. Children with symptomatic vitamin E deficiency should be treated with 400 mg/d orally of water-miscible esters; alternatively, 2 mg/kg per d may be administered intramuscularly. Vitamin E in high doses may protect against oxygen-induced retrolental fibroplasia and bronchopulmonary dysplasia, as well as intraventricular hemorrhage of prematurity.

Posterior Ischemic Optic Neuropathy This is an infrequent cause of acute visual loss, induced by the combination of severe anemia and hypotension. Cases have been reported after major blood loss during surgery, exsanguinating trauma, gastrointestinal bleeding, and renal dialysis. The fundus usually appears normal, although optic disc swelling develops if the process extends far enough anteriorly. Vision can be salvaged in some patients by prompt blood transfusion and reversal of hypotension. Optic Neuritis This is a common inflammatory disease of the optic nerve.

The contribution of genetics to presbycusis (see below) is also becoming better understood. In addition to GJB2, several other nonsyndromic genes are associated with hearing loss that progresses with age. Sensitivity to aminoglycoside ototoxicity can be maternally transmitted through a mitochondrial mutation. Susceptibility to noise-induced hearing loss may also be genetically determined. There are 400 syndromic forms of hearing loss.
