Isopropyl β-D thiogalactopyranoside

Bougainvillea antiviral protein (BAP) is one among a class of the ribosomal inactivating proteins isolated from Bougainvillea spectabilis willd. Truncated version of the BAP gene was cloned and expressed in a prokaryotic vector to abolish its cytotoxicity. RNA was isolated from mature leaves of Bougainvillea and the full length cDNA was amplified by reverse transcription-PCR using template mRNA. This full length cDNA of size 756 bp was amplified using the proofreading polymerase (Q5 polymerase) and end to end gene specific primers for removal of C-terminal, the amplicon was cloned in pJET1.

The present study aims at evaluating the efficacy of two rapid tools, IgM dot ELISA and Latex Agglutination Test employing recombinant LipL32 as antigen in diagnosing acute leptospirosis in dogs in Kerala, a disease endemic area. One hundred and five serum samples were collected from dogs presented at Veterinary Hospitals attached to Kerala Veterinary and Animal Sciences University during the period from September 2014 to May 2016. The gene encoding ribulokinase araB from Anoxybacillus kestanbolensis AC26Sari was cloned and sequenced. The recombinant protein was expressed in Escherichia coli BL21 under the control of isopropyl-β-D-thiogalactopyranoside-inducible T7 promoter.

A porcine fat mass and obesity-associated gene (pFTO) was cloned into the expression vector pET30a(+) and heterologously expressed in Escherichia coli. The effects of isopropyl β-D thiogalactopyranoside (IPTG) concentration and induction time on the expression of recombinant pFTO were assessed.
