Triple negative breast cancer (TNBC) is a distinct subtype of breast cancer burdened with a dismal prognosis due to the lack of effective therapeutic agents. Receptors for LHRH (luteinizing hormone-releasing hormone) can be successfully targeted with AEZS-108 [AN-152], an analog of LHRH conjugated to doxorubicin.

Luteinizing hormone-releasing hormone receptor (LHRHr) represents a promising therapeutic target for treating sex hormone-dependent tumors. We coupled cecropin B, an antimicrobial peptide, to LHRH’, a form of LHRH modified at carboxyl-terminal residues 4–10, which binds to LHRHr without interfering with luteinizing hormone (LH) and follicle-stimulating hormone (FSH) secretion.

Although brucea javanica oil liposomes (BJOLs) have been used clinically to treat ovarian cancer, its clinical efficacy is often limited by systemic side effects due to non-specific distribution. Luteinizing hormone releasing hormone receptor (LHRHR) is overexpressed in most ovarian cancers but negligibly expressed in most of the other visceral organs.

We have shown that hormone-induced downregulation of luteinizing hor-mone receptor (LHR) in the ovary is post-transcriptionally regulated by an mRNA binding protein. This protein, later identified as mevalonate kinase (MVK), binds to the coding region of LHR mRNA, suppresses its transla-tion, and the resulting ribonucleoprotein complex is targeted for degrada-tion.

Gonadotropin releasing hormone receptor (GnRHR) is secreted from hypothalamus and delivered to the anterior pituitary via the hypophyseal portal circulation where it binds to the GnRHR on the surface of gonadotropes triggering the synthesis and secretion of the gonadotropins, follicle stimulating hormone (FSH) and luteinizing hormone (LH). In the female, LH stimulates the production of androgens by the thecal cells that surround the growing ovarian follicle.

Gonadotropin-releasing hormone antagonists (GnRH-nt) available for clinical use are GnRH molecules with amino acid modifications in positions 1, 2, 3, 6, and 10. They are not associated with the histaminic-release effects of previous compounds (1). These compounds immediately block GnRH receptor in a competitive fashion (2). They decrease the luteinizing hormone (LH) and follicle-stimulating hormone (FSH) secretion within a period of eight hours. Inhibition of LH secretion is more important than FSH.
