Molecular characterisation

This textbook represents an attempt to present a relatively brief overview of Materials Science, the anticipated readership being students of structural and mechanical engineering. It is in two sections – the first characterising engineering materials, the second considering structure–property relationships. Emphasis is thus placed on the relationship between structure and properties of materials, starting with the concept of ‘structure’ at three levels - crystal structure, microstructure, and molecular structure.

Mutation acquisition is a major mechanism of bacterial antibiotic resistance that remains insufficiently characterised. Here we present RM-seq, a new amplicon-based deep sequencing workflow based on a molecular barcoding technique adapted from Low Error Amplicon sequencing (LEA-seq).

High-grade serous tubo-ovarian cancer (HGSTOC) is characterised by extensive inter- and intratumour heterogeneity, resulting in persistent therapeutic resistance and poor disease outcome. Molecular subtype classification based on bulk RNA sequencing facilitates a more accurate characterisation of this heterogeneity, but the lack of strong prognostic or predictive correlations with these subtypes currently hinders their clinical implementation.

The association between microbes and cancer has been reported repeatedly; however, it is not clear if molecular tumour properties are connected to specific microbial colonisation patterns. This is due mainly to the current technical and analytical strategy limitations to characterise tumour-associated bacteria.

Ebook "UV-B radiation and plant life: Molecular biology to ecology" deals with large-scale impacts but also how UV-B affects plants at the molecular level is also fascinating, and the UV-B photoreceptor has only recently been characterised. While UV-B radiation can be damaging, it also has a more positive role in plant photomorphogenesis. Consequently UV-B treatments are being developed as innovative approaches to improve horticulture. This book is a timely synthesis of what we know and need to know about UV-B radiation and plants.

The genus Scutellonema consists of about 50 species that are phytophagous (ecto- and partially endo-parasites). Among species of the genus Scutellonema, Scutellonema brachyurum was reported to be one of the three most damaging ones, causing serious symptoms to numerous host plants worldwide.

This project investigates the use of free radical initiators in the recycling of post consumer recycled high density polyethylene using reactive extrusion. Both molecular and rheological characterisation of recycled and reprocessed materials was carried out and this was followed by tensile testing of the modified materials to satisfy end use applications such as packaging and drainage piping.

The increase in new cases of drug resistance to first-line drugs such as ethambutol (ETB) makes it necessary to develop improvements for antituberculosis drugs. A new method for improving the bioavailability of active pharmaceutical ingredient (API) was investigated for preventing drug resistance and side-effects of antituberculosis drugs. In this study, an antituberculosis drug delivery system using β-cyclodextrin (β-CD) as the supramolecular carrier of ETB was prepared using the kneading method.

The study "Characterization of grapevine (V. vinifera L.) varieties grown in Yozgat province (Turkey) by simple sequence repeat (SSR) markers" was conducted to characterise 50 grape varieties grown in Yozgat by molecular methods. Molecular definitions were realized using 9 simple sequence repeat (SSR) primers (VVS2, VVMD5, VVMD7, VVMD24, VVMD27, VVMD28, VVMD31, VrZAG62, and VrZAG79), including six microsatellite loci used for grapevine variety collections by the European Union funded project (GENRES 081), accepted as the minimum standard set (core set) by the world.

A set of mutant pea lines carrying induced mutations within the major seed-expressed starch-branching enzyme gene has been characterised at the molecular, chemical and agronomic levels. Eight of the induced mutations, three of which predicted a premature stop codon, were compared with the naturally occurring starch-branching enzyme mutation within the same genetic background.

In the present work a galactomannan extract of low protein residue (< 1.3 % wt dry basis) was isolated from alfalfa (Medicago sativa L.) seed endosperm meal. The alfalfa gum (AAG) comprised primarily mannose and galactose at a ratio of 1.18:1, had a molecular weight of 2 × 106 Da and a radius of gyration of 48.7 nm.

Many materials used in a wide range of fields consist of polymers that feature great structural complexity. One particularly suitable technique for characterising these complex polymers, that often feature correlated distributions in e.g. microstructure, chemical composition, or molecular weight, is comprehensive two-dimensional liquid chromatography (LC × LC).

Antimicrobial resistance (AMR) is an increasing global health concern reducing options for therapy of infections and also for perioperative prophylaxis. Many Enterobacteriaceae cannot be treated anymore with third generation cephalosporins (3GC) due to the production of certain 3GC hydrolysing enzymes (extended spectrum beta-lactamases, ESBLs).

The synthesis and characterisation including 1 H and 13 C NMR spectroscopy of a Mannich base 1,4-di-(5- acetamido-2-hydroxybenzyl)piperazine is herein reported. The Mannich reaction leading to the formation of the ligand took place at both ends of piperazine. Four metal complexes [Cu(II) and Fe(III)] including those bearing thiocyanate groups (SCN–) in N- and S-bonding modes have been studied for their abilities to mimic catecholase oxidase. All the metal complexes are catalytically active with the highest turnover rate (k cat) recorded for complex 3.

The outcome of cervical artificial insemination (AI) with frozen-thawed semen in sheep is limited by the inability of sperm to traverse the cervix of some ewe breeds. Previous research has demonstrated that cervical sperm transport is dependent on ewe breed, as sperm can traverse the cervix in greater numbers in some higher fertility ewe breeds. However, the molecular mechanisms underlying ewe breed differences in sperm transport through the cervix remain unknown.
