Cytochrome c is widely distributed in bacterial species, from mesophiles to thermophiles, and is one of the best-characterized redox proteins in terms of biogenesis, folding, structure, function, and evolution. Experimental molecular biology techniques (gene cloning and expression) have become applicable to cytochrome c, enabling its engineering and manipulation. Heterologous expression systems for cytochromes c in bacteria, for use in mutagenesis studies, have been established by extensive investigation of the biological process by which the functional structure is formed. ...

In this review, we summarize recent progress in studying three main classes of prenyltransferases: (a) isoprenyl pyrophosphate synthases (IPPSs), which catalyze chain elongation of allylic pyrophosphate substrates via consecutive condensation reactions with isopentenyl pyrophosphate (IPP) to generate linear polymers with defined chain lengths; (b) protein prenyltransferases, which catalyze the transfer of an isoprenyl pyrophosphate (e.g. farnesyl pyrophosphate) to a protein or a peptide; (c) prenyltransferases, which catalyze the cyclization of isoprenyl pyrophosphates.

PSA (prostate-specific antigen), the most useful serum marker for prostate cancer, is encoded by thehKLK3gene and is present in the serumas amixture of several molecular species. This work was performed to identify the hKLK3 transcripts in order to determine how many proteins resembling PSA are synthesized from thehKLK3gene and secreted in blood. Combined Northern blotting, molecular cloninganddatabase searching showed that thehKLK3gene produces at least 15 transcripts ranging in size from 0.7 to 6.1 kb. Polysomal distribution analysis revealed that the transcripts shorter than 3.

Serine⁄threonine kinase 33 (STK33⁄Stk33) is a recently discovered gene whose inferred amino acid sequence translation displays characters typical for a calcium⁄calmodulin dependent kinase (CAMK). In this study we ana-lysed the STK33⁄Stk33RNA and protein distribution and the localization of the protein. TheSTK33⁄Stk33expression pattern resembles those of some related members of the CAMK group.STK33⁄Stk33displays a non-ubiquitous and, in most tissues, low level of expression.

Poly(b-L-malate) (PMLA) has been reported as an uncon-ventional, physiologically important biopolymer in plasmo-dia of myxomycetes, and has been proposed to function in the storage and transport of nuclear proteins by mimicking the phospho(deoxy)ribose backbone of nucleic acids. It is distributed in the cytoplasm and especially in the nuclei of these giant, multinucleate cells. We report here for the first time an increase in growth rate and a shortening of the cell cycle after the injection of purified PMLA. ...

Complexation of the small serine protease inhibitorSchistocerca gregaria chymotrypsin inhibitor (SGCI), a member of the pacifastin inhibitor family, with bovine chymotrypsin was followed by NMR spectroscopy. 1 H– 15 N correlation (HSQC) spectra of the inhibitor with increasing amounts of the enzyme reveal tight and specific binding in agreement with biochemical data.

Antimicrobial peptides are an abundant group of proteinaceous com-pounds widely produced in the plant kingdom. Among them, thec-thionin family, also known as plant defensins, represents one typical family and comprises low molecular mass cysteine-rich proteins, usually cationic and distributed in different plant tissues.

A chilly, but still very attractive, Asilomar hosted the seventh edition of the Critical Assessment of Protein Structure Prediction Experiment (CASP) [1]. A few months ago, 96 protein sequences of unknown, but soon to be determined structure, were distributed to more than 250 groups, whose task was to predict their structure and a number of other fea-tures, such as their domain boundaries, the presence of disordered regions, contacts between specific amino acids and their molecular function.

Glutathione (GSH) plays a key role in defense against oxidative stress. The availability of GSH is ensured in tissues by systems devoted to its mainte-nance in the reduced state and by the flux of GSH and cysteine between sites of biosynthesis and sites of utilization. Little is known about the effect of oxidative stress on the distribution of low-molecular-mass thiols and their exchange rate between tissues.

Insulin like growth factor-1 (IGF-1) is established as an anabolic factor that can induce skeletal muscle growth by activating the phosphoinosi-tide 3-kinase⁄Akt⁄mammalian target of rapamycin (mTOR) pathway. Although this signaling pathway has been the subject of much study, the molecular mechanisms linking IGF-1 binding to mTOR activation remain poorly defined in muscle.

Although numerous molecular changes have been reported in VSCC, biomarkers of clinical relevance are still lacking. On the other hand, there is emerging evidence on the use of mtDNA as a diagnostic tool in oncology.

Sindbis virus (SV) is the prototype of alphaviruses which are a group of widely distributed human and animal pathogens. Heterogeneous nuclear ribonucleoprotein (hnRNP) A1 is an RNA-binding protein that shuttles between the nucleus and the cytoplasm. Our recent studies found that hnRNP A1 relocates from nucleus to cytoplasm in Sindbis virus (SV)-infected cells. hnRNP A1 binds to the 5′ UTR of SV RNA and facilitates the viral RNA replication and translation.
