Ebook "Studies on periodontal disease (Oxidative stress in applied basic research and clinical practice series)" comprise four parts. Part 1 provides basic sciences from the general to the particular. The findings of cell culture and animal models supply the mechanism between periodontal disease and oxidative stress. In part 2, human clinical studies are mainly addressed. The effects of periodontal therapy on circulating oxidative stress are also discussed. The relationships between periodontal disease and systemic diseases are examined in part 3.

Attention deficit hyperactivity disorder (ADHD) is characterized by symptoms of inattention, excessive motor activity and impulsivity detected mostly during childhood. These traits are known to be controlled by monoamine neurotransmitters, chiefly dopamine, serotonin and norepinephrine.

Unsaturated ketone derivatives are known as monoamine oxidase B (MAO-B) inhibitors, a potential drug target for Parkinson’s disease. Here, molecular modeling studies, including 2D-QSAR, ADMET prediction, molecular docking, and MD simulation, were performed on a new series of MAO-B inhibitors. The objective is to identify new MAO-B inhibitors with high inhibitory efficacy. The developed 2D-QSAR model was based on the descriptors of MOE software.

New 3-aryl-6-methylpyrimido[4,5-c]pyridazine-5,7(6H,8H)-diones and 3-aryl-6-ethyl-7-thioxo-7,8-dihydropyrimido[4,5-c]pyridazin-5(6H)-ones were efficiently synthesized via a regiospecific one-pot reaction of N-methylbarbituric acid and N -ethyl-2-thiobarbituric acid with various arylglyoxal monohydrates in the presence of hydrazine dihydrochloride in ethanol at 50◦C. The target compounds were obtained in high yields and were regioisomerically pure after recrystallization. These new heterocycles may act as potential MAO B inhibitors.

A series of new thiazole-hydrazones (3a–3n) were synthesized, characterized, and screened for their hMAOA and hMAO-B inhibitory activity by an in vitro fluorometric method. Selectivity indexes (SIs) were expressed as IC50 (MAO-A) / IC50 (MAO-B). Compound 3f showed promising hMAO-A inhibition with an IC50 value of 1.20 µM and displayed a very significant SI of 0.04 towards hMAO-A. The mechanism of hMAO-A inhibition was investigated by enzyme kinetics using Lineweaver–Burk graphics.

The two forms of monoamine oxidase, monoamine oxidase A and mono-amine oxidase B, have been associated with imidazoline-binding sites (type 2). Imidazoline ligands saturate the imidazoline-binding sites at nanomolar concentrations, but inhibit monoamine oxidase activity only at micromolar concentrations, suggesting two different binding sites [Ozaita A, Olmos G, Boronat MA, Lizcano JM, Unzeta M & Garcı´a-Sevilla JA (1997) Br J Pharmacol121,901–912].

A common feature of all the proposed mechanisms for monoamine oxidase is the initiation of catalysis with the deprotonated form of the amine sub-strate in the enzyme–substrate complex. However, recent steady-state kinetic studies on the pH dependence of monoamine oxidase led to the sug-gestion that it is the protonated form of the amine substrate that binds to the enzyme.

The advantages which have been derived from the caution with which hypothetical statements are admitted, are in no instance more obvious than in those sciences which more particularly belong to the healing art. It therefore is necessary, that some conciliatory explanation should be offered for the present publication: in which, it is acknowledged, that mere conjecture takes the place of experiment; and, that analogy is the substitute for anatomical examination, the only sure foundation for pathological knowledge.

James Parkinson described Parkinson’s disease in his memorable Essay on the Shaking Palsy in 1817. Since then, and particularly in recent years, there has been tremendous progress in our understanding of this complex and fascinating neurological disorder. Briefly, we have learned that it is not only manifest by motor symptoms but also that there is a whole range of non-motor features, including autonomic, psychiatric, cognitive and sensory impairments. We now know how to distinguish better clinically between Parkinson’s disease and the various parkinsonian syndromes.
