Ebook "Adult stem cell therapies: Alternatives to plasticity" explores various techniques for tissue and organ regeneration using stem cells isolated from adult tissues. It discusses alternative explanations of stem cell plasticity as well as current clinical results with adult stem cell therapies. It examines the presence of potential pluripotent stem cells in adult tissues, paracrine effects of stem cell therapies, and involvement of exosomes and microparticles into observed phenomena.

The source of multipotent stromal cells (MSC) can have a significant influence on the health and expansion capacity of the cells. As the applications for allogeneic MSCs in the treatment of feline diseases increase, the location of the initial donor tissue must be analyzed.

Single-cell transcriptomics allows researchers to investigate complex communities of heterogeneous cells. It can be applied to stem cells and their descendants in order to chart the progression from multipotent progenitors to fully differentiated cells. While a variety of statistical and computational methods have been proposed for inferring cell lineages, the problem of accurately characterizing multiple branching lineages remains difficult to solve.

Adipose tissue-derived stem cells (ADSCs), a type of mesenchymal stem cells (MSCs), have great potential as therapeutic agents in regenerative medicine. Numerous animal studies have documented the multipotency of ADSCs, showing their capabilities to differentiate into tissues such as muscle, bone, cartilage, and tendon.

Subcutaneous adipose tissue represents an abundant source of multipotent adult stem cells named as Adipose-derived stem cells (ADSCs). With a cell sheet approach, ADSCs survive longer, and can be delivered in large quantities. We investigated whether intra-articular ADSC sheets attenuated osteoarthritis (OA) progression in a rabbit anterior cruciate ligament transection (ACLT) model.

The presence of tumor supporting cells in various cancer, including in hepatocellular carcinoma (HCC), has become an important target in the study of carcinogenesis. The cancer-associated fibroblast (CAF), one of the most important cellular components in the cancer stroma, might contribute to the progression of the disease due to its plasticity, a behavior of the stem cells. In this study, we investigate the significance of the CAF and its role in the HCC progression and metastasis.

Mesenchymal stem cells are multipotent/pluripotent stromal cells that can differentiate into a diversity of cell categories depending upon the micro-environment to that they are exposed. The multipotent differentiation potential of MSCs is generally towards a constrained sum of cell types familiar to the tissue of origin and maintains the capacity for self-renewal. To assess the differentiation potential of MSCs to skeletal myocytes a male mongrel dog of 3 years was used to carry out the research.

Hair follicle is an ideal system for studying the biology and regulation of adult stem cells. Multipotent stemness of the canine hair follicle stem cells (cHFSCs) were detected by immunostaining the cells with positive HFSC surface markers like Anti-cytokeratin-15 (CK15) and CD200. In this study, canine hair follicular cells were cultured in Williams’E media supplemented with growth factors.

For cell-based bone tissue engineering, various tissues derived cells are utilized since osteogenic cells can be harvested from bone marrow, periosteum, and adipose tissue, though recent studies indicate that bone marrow stromal cells (BMSCs, bone marrow derived multipotent mesenchymal stromal cells, or mesenchymal stem cells) are the most reliable cell source because of their superior osteogenic ability (Hayashi et al., 2008). However, it is difficult to obtain adequate numbers of transplantable BMSCs from bone marrow aspirates, as they are rare in the bone marrow (less than 0.

Islet transplantation provides a promising cure for Type 1 diabetes; however it is limited by a shortage of pancreas donors. Bone marrow-derived multipotent mesenchymal stem cells (MSCs) offer renewable cells for generating insulin-producing cells (IPCs). Methods: We used a four-stage differentiation protocol, containing neuronal differentiation and IPC-conversion stages, and combined with pellet suspension culture to induce IPC differentiation.

Hierarchy of hematopoietic differentiation. Stem cells are multipotent cells that are the source of all descendant cells and have the capacity to provide either long-term (measured in years) or short-term (measured in months) cell production. Progenitor cells have a more limited spectrum of cells they can produce and are generally a short-lived, highly proliferative population also known as transient amplifying cells. Precursor cells are cells committed to a single blood cell lineage but with a continued ability to proliferate; they do not have all the features of a fully mature cell.

Nuclear Reprogramming Development naturally progresses from totipotent fertilized eggs to pluripotent epiblast cells, to multipotent cells, and finally to terminally differentiated cells. According to Waddington's epigenetic landscape, this is analogous to a ball moving down a slope. The reversal of the terminally differentiated cells to totipotent or pluripotent cells (called nuclear reprogramming) can thus be seen as an uphill gradient that never occurs in normal conditions.
