Pikovskaya medium

In the present study ten different Bacillus isolates were isolated from various soil samples and their identification was performed on the basis of Gram staining, colony morphology and biochemical tests. Gram positive rod shaped bacteria showing large, circular, opaque colonies having white or greyish white appearance were preserved on Nutrient agar slants for further identification. All isolates showed positive results for Catalase, Urease, Oxidase and motility biochemical tests.

A field experiment was conducted with maize-groundnut cropping sequence. Rock phosphate and its combination with SSP were used as nutrient sources during Rabi 2016- 17 (maize taken up in Kharif 2016). The study was carried out in the Central Farm, OUAT with the help of a field experiment laid out in Randomized Block Design with seven treatment T1 Control, T2 100% P(RP),T3 100% P(SSP), T4 75% P(RP) + 25% P(SSP), T5 50% P(RP) + 50% P(SSP), T6 25% P(RP) + 75% P(SSP) and T7 100% P(SSP) + Lime @ 0.2 LR and replicated in thrice.

In the present study endophytic bacteria were isolated from Aegle marmelos and were screened for phosphate solubilizing activity. The isolate AP01 showed the ability to solubilize inorganic tricalcium phosphate and thus was characterized. Phenotypic, biochemical and genotypic analysis revealed, the isolate to be Kosakonia cowanii, a gram –ve proteobacterium of family enterobacteriaceae with accession no. MK834804, of the three media viz.

A total of forty rhizosphere soil samples were collected from different locations of Bengaluru urban district, Karnataka and used for isolation of PGPRs by standard dilution plate method. Seventy bacterial isolates were selected, purified and were examined for morphological characters like colony morphology, cell shape, Gram reaction. Isolates were diverse in their morphology ranging from dull to milky white, rough to smooth margins, viscous, small to large, non-spreading and spreading.

Plants acquire phosphorus from soil solution as phosphate anion. It is the least mobile element in plant and soil contrary to other macronutrients. In the present study phosphate solubilizing bacteria were isolated from rhizosphere of four different crops on Pikovskaya’s agar medium. The potential isolate was selected on the basis of reduction in pH of media and formation of halo zone. Its biochemical study was conducted which conclude that the isolate belong to Bacillus sp.
