Ras gene mutation and/or overexpression are drivers in the progression of cancers, including colorectal cancer. Blocking the Ras signaling has become a significant strategy for cancer therapy. Previously, we constructed a recombinant scFv, RGD-p21Ras-scFv by linking RGD membrane-penetrating peptide gene with the anti-p21Ras scFv gene.

The Phenomenon of codon usage bias exists in the genomes of prokaryotes and eukaryotes. The codon usage pattern is afected by environmental factors, base mutation, gene fow and gene expression level, among which natural selection and mutation pressure are the main factors. The study of codon preference is an efective method to analyze the source of evolutionary driving forces in organisms.

Flowers of safflower (Carthamus tinctorius L.) have been widely used in traditional Chinese medicine because of their ability to improve cerebral blood flow. The major active ingredients in safflower are flavonoids. To date, few flavonoid biosynthesis genes have been cloned from this species. In our previous research, four isoform chalcone isomerase genes were screened. In this study, a new chalcone isomerase gene (designated as CtCHI-N) was cloned from a safflower cDNA library. A 696-bp cDNA for CtCHI-N was cloned, and its sequence was subsequently analyzed.

Rheum palmatum has been widely developed in traditional Chinese medicine. One of the major active compounds in R. palmatum is flavonoids. Until now, few genes involved in flavonoid biosynthesis have been isolated from this medicinal plant. In this study, a new chalcone isomerase gene (RpCHI) was cloned from an RNA-seq library of R. palmatum. The 714bp RpCHI cDNA was cloned and sequenced.

. In this study, the VEGF121 gene was cloned and expressed in a prokaryotic expression system (Escherichia coli). The recombinant VEGF121 was encapsulated with PLA microparticles and studied in vitro and ex ovo for the sustained release mechanism. The PLA-VEGF microparticles and the recombinant VEGF121 were explored for their bioactivity in human umbilical vein endothelial cells (HUVEC).

Heat shock transcription factor (Hsfs) is widely found in eukaryotes and prokaryotes. Hsfs can not only help organisms resist high temperature, but also participate in the regulation of plant growth and development (such as involved in the regulation of seed maturity and affects the root length of plants).

Histone acetylation is a ubiquitous and reversible post-translational modification in eukaryotes and prokaryotes that is co-regulated by histone acetyltransferase (HAT) and histone deacetylase (HDAC). HAT activity is important for the modification of chromatin structure in eukaryotic cells, affecting gene transcription and thereby playing a crucial regulatory role in plant development.

RNA-seq and its variant differential RNA-seq (dRNA-seq) are today routine methods for transcriptome analysis in bacteria. While expression profiling and transcriptional start site prediction are standard tasks today, the problem of identifying transcriptional units in a genome-wide fashion is still not solved for prokaryotic systems.

The σB protein of avian reovirus (ARV) is diagnostically important protein for development of an ARV-specific enzyme-linked immunosorbent assay (ELISA) and for which production of an antigenic recombinant σB protein in abundance is a prerequisite. In the present study, we successfully employed the prokaryotic system for expression of antigenic σB protein of ARV.

Human mitochondrial peptide deformylase (PDF) has been proposed as a novel cancer therapeutic target. However, very little is known about its expression and regulation in human tissues. The purpose of this study was to characterize the expression pattern of PDF in cancerous tissues and to identify mechanisms that regulate its expression.

Therefore in this study immunodominant region in TP53 gene was amplified and cloned in prokaryotic expression vector. Further the conditions were optimized for the expression of the recombinant protein. The recombinant protein was purified using Ni-NTA affinity chromatography. The hyperimmune sera were raised in rabbits against the recombinant protein. In the hyperimmune sera, antibodies specific to TP53 were detected by Indirect ELISA, confirming the serological reactivity of the immunodominant region.

Heat shock proteins (HSPs) usually act as molecular chaperones to prevent proteins from being denatured in response to high temperature and other extreme conditions. These are highly conserved from prokaryotes to eukaryotes. In this study, we analysed the differential expression pattern of hsp21 and hsp70 transcript during thermal stress in tropical tasar silkworm. We found expression of hsp21 and hsp70 were upregulated ~2.5 and 4 folds respectively in the temperature treated larvae in comparison to the control larvae.

Cloning and expression of recombinant VP2 capsid protein gene of canine parvovirus in E. coli System
In the present study, new CPV-2a field strain (KLD3) isolated in cell culture was selected and the whole CPV VP2 gene was used for the expression in the E. coli expression system. Prokaryote expressing monocistronic DNA cassette containing open reading frame of whole CPV capsid gene (VP2) downstream of T7 promoter was synthesized. Analysis of the expression in E. coli cells showed the presence of capsid protein. Recombinant capsid protein showed immunoreactivity similar to the whole CPV virus antigen, when reacted with polyclonal antibodies against the whole CPV virus particles.

Bluetongue is an arthropod-borne, non-contagious viral disease of domestic and wild ruminants. In the present study, truncated VP2 protein of BTV serotype 23 (BTV-23) was expressed in a prokaryotic expression system and assessed for its potential use as a candidate for the development of serotype-specific BTV diagnostics assay. The expression of the VP2 protein was carried out in E. coli BL21 Codon Plus (DE3) cells using the pET32a expression vector.
