Determinants for desensitization and sequestration of G protein-coupled receptors often contain serine or threonine residues located in their C-ter-mini. The sequence context, however, in which these residues have to appear, and the receptor specificity of these motifs are largely unknown.

The building blocks of most signal transduction pathways are pairs of enzymes, such as kinases and phosphatases, that control the activity of pro-tein targets by covalent modification. It has previously been shown [Gold-beter A & Koshland DE (1981)Proc Natl Acad Sci USA78, 6840–6844] that these systems can be highly sensitive to changes in stimuli if their cata-lysing enzymes are saturated with their target protein substrates.

To elucidate the architectural principle of protein structure, we focused on sequestration from solvent, which is a common characteristic of folding and self-associative precipitation. Because protein solubility can be regarded as a basis for the potential ability to sequester from solvent, we assume that poorly soluble proteins tend not only to precipitate, but also to form solution structures.

The role of non-B DNA in the function and stability of genomes has become generally appreciated in recent years. It is now evident that DNA also encodes for spatial structures that are involved in gene regulation, replication and recombination.

The ability of mitochondria to sequester and retain divalent cations in the form of precipitates consisting of organic and inorganic moieties has been known for decades. Of these cations, Ca 2+ has emerged as a major player in both signal transduction and cell death mechanisms, and, as a conse-quence, the importance of mitochondria in these processes was soon recog-nized.

Calcium is an extraordinarily versatile signaling ion, encoding cellular responses to a wide variety of external stimuli. In neurons, mitochondria can accumulate enormous amounts of calcium, with the consequence that mitochondrial calcium uptake, sequestration and release play pivotal roles in orchestrating calcium-dependent responses as diverse as gene transcrip-tion and cell death.
