The interaction of blood coagulation factor X and its Gla-containing fragments with negatively charged phospholipid membranes composed of 25 mol% phosphatidylserine (PtdSer) and 75 mol% phosphatidylcholine (PtdCho) was studied by surface plasmon resonance. The binding to 100 mol% PtdCho membranes was negligible. The calcium dependence in the membrane binding was evaluated for intact bovine factor X (factor X) and the fragment containing the Gla-domain and the N-terminal EGF (epidermal growth factor)-like domain, Gla–EGFN, from factor X....

Division of Medicinal and Natural Products Chemistry, Department of Chemistry and Department of Chemical Engineering, University of Iowa, USA; 2Department of Autoimmunology, Statens Serum Institut, Copenhagen, Denmark Synthetic peptides based on amino-acid residues 27–38 of human serum amyloid P component represent a novel type of heparin binders as they do not contain clusters of basic amino acids or other known features associated with protein or peptide heparin binding.

The interaction between the c11ring and thececomplex, forming the rotor of theIlyobacter tartaricus ATP synthase, was probed by surface plasmon resonance spectroscopy andin vitro reconstitution analysis. The results pro-vide, for the first time, a direct and quantitative assessment of the stability of the rotor. The data indicated very tight binding between the c11ring and the cecomplex, with an apparent Kd value of approximately 7.4 nm.

The catalytic activity of phenylalanine hydroxylase (PAH, phenylalanine 4-monooxygenase EC 1.14.16.1) is regulated by three main mechanisms, i.e. substrate (L-phenylalanine, L-Phe) activation, pterin cofactor inhibition and phos-phorylation of a single serine (Ser16) residue. To address the molecular basis for the inhibition by the natural cofactor (6R)-L-erythro-5,6,7,8-tetrahydrobiopterin, its effects on the recombinant tetrameric human enzyme (wt-hPAH) was studied using three different conformational probes, i.e.

A library of random peptide sequences was used to select peptides that inhibit an anti-idiotypic catalytic Ig, immuno-globulin (IgG) 9G4H9, withab-lactamase-like activity. This library displays cyclic heptapeptides on the surface of bac-teriophagesand representsacollectionofupto4.5·10 9 peptides. The first selection step aimed at enriching the lib-rary in species that bind to the whole Ig molecule. The sec-ond stepwas todiscriminate peptides that bind topart of the molecule other than the active site. Selected peptides were then screened by surface plasmon resonance analysis.

Novel antithrombin molecules were identified from the ixodidae tick, Haemaphysalis longicornis. These molecules, named madanin 1 and 2, are 7-kDa proteins and show no significant similarities to any previously identified proteins. Assays using human plasma showed that madanin 1 and 2 dose-dependently prolonged both activated partial throm-boplastin time and prothrombin time, indicating that they inhibit both the intrinsic and extrinsic pathways. Direct binding assay by surface plasmon resonance measurement demonstrated that madanin 1 and 2 specifically interacted with thrombin....

Thrombomodulin (TM) slows down the interaction rate between thrombin and plasminogen activator inhibitor 1 (PAI-1). We now show that the 12-fold reduced inhibition rate in the presence ofTM does not result from an altered distribution between PAI-1 cleavage and irreversible com-plex formation. Surface plasmon resonance (SPR) revealed an over 200-fold reduced affinity of TM for thrombin-VR1 tPA as compared to thrombin, demonstrating the importance ofthe VR1 loop in the interaction ofthrombin withbothTMandPAI-1....

The enzymes pyruvate decarboxylase (E1) and dihydro-lipoyl dehydrogenase (E3) bind tightly but in a mutually exclusive manner to the peripheral subunit-binding domain (PSBD) of dihydrolipoyl acetyltransferase in the pyruvate dehydrogenase multienzyme complex of Bacillus stearo-thermophilus.

Báo cáo khoa học: Mapping the functional domains of human transcobalamin using monoclonal antibodies
Recombinant human transcobalamin (TC) was probed with 17 monoclonal antibodies (mAbs), using surface plasmon resonance measurements. These experiments identified five distinct epitope clusters on the surface of holo-TC. Western blot analysis of the CNBr cleavage fragments of TC allowed us to distribute the epitopes between two regions, which spanned either the second quarter of the TC sequence GQLA…TAAM(103–198) or the C-ter-minal peptide LEPA…

The phosphoproteins HPrSerP and CrhP are the main effectors for CcpA-mediated carbon catabolite regulation (CCR) inBacillus subtilis. Complexes of CcpA with HPrSerP or CrhP regulate genes by binding to the catabolite responsive elements (cre). We present a quantitative analysis of HPrSerP and CrhP interaction with CcpA by surface plasmon resonance (SPR) revealing small and similar equilibrium constants of 4.8 ± 0.4 lm for HPrSerP–CcpA and 19.1 ± 2.5 lmfor CrhP–CcpA complex dissoci-ation.

I C , an endogenous cytoplasmic inhibitor of vacuolar carboxypeptidase Y in the yeast Saccharomyces cerevisiae, is classified as a member of the phos-phatidylethanolamine-binding protein family. The binding of I C to phos-pholipid membranes was first analyzed using a liposome-binding assay and by surface plasmon resonance measurements, which revealed that the affin-

The topoisomerase I (TopI) reaction intermediate consists of an enzyme covalently linked to a nicked DNA molecule, known as a TopI-DNA complex, that can be trapped by inhibitors and results in failure of re-ligation. Attempts at new derivative designs for TopI inhibition are enthusiastically being pursued, and TopI inhibitors were developed for a variety of applications. Surface plasmon resonance (SPR) was recently used in TopI-inhibition studies.
