The concept of coworking caters to freelancers and entrepreneurs who not only seek a shared office space but also be part of a socially bound group with similar ideas and goals. The aim of providing a space that fosters collaboration, sense of community, enhances creativity and sustains communication within a diverse group of independent professionals. Whilst the concept has flourished over the span of 10 years, little is known of the participants’ experiences within coworking spaces and whether such spaces do deliver what it promises.

This dissertation has been developed to understand how coworkers construct their identity within the Hive. With literature supporting the notion of identity construction such as identity struggle, organizational identification, identification of optimal balances between self and professional identities within traditional bureaucratic organizations, the aim of this dissertation seeks to determine how identity work is being constructed within a coworking space in a single case study.

An oxidosqualene cyclase cDNA, LcIMS1, was isolated from cultured cells of Luffa cylindrica Roem. by heterologous hybridization with cDNA of Glycyrrhiza glabra b-amyrin synthase. Expression of LcIMS1 in yeast lacking endogenous oxidosqualene cyclase activity resulted in the accumulation of isomultiflorenol, a triterpene. This is consistent with LcIMS1 encoding isomultiflorenol synthase, an oxidosqualene cyclase involved in bryonolic acid biosynthesis in cultured Luffa cells.

From the antennae of the moth Mamestra brassicae, we have identified a lepidopteran G protein a subunit belonging to the Gq family, through immunological detection in crude antennal extract and antennal primary cell cultures, followed by molecular cloning. The complete cDNA sequence (1540 bp) contains an open reading frame encoding a protein of 353 amino acids.

We have isolated and characterized a second [Fe]-hydro-genasegene fromthegreenalga,Chlamydomonas reinhardtii. TheHydA2gene encodes a protein of 505 amino acids that is 74% similar and 68% identical to the known HydA1 hydrogenase fromC. reinhardtii.HydA2 contains all the conserved residues and motifs found in the catalytic core of the family of [Fe]-hydrogenases. We demonstrate that both theHydA1and theHydA2transcripts are expressed upon anaerobic induction, achieved either by neutral gas purging or by sulfur deprivation of the cultures. ...

We have cloned, over-expressed and purified enolase from Plasmodium falciparumstrain NF54 in Escherichia coliin active form, as an N-terminal His6-tagged protein. The sequence of the cloned enolase from the NF54 strain is identical to that of strain 3D7 used in full genome sequen-cing. The recombinant enolase (r-Pfen) could be obtained in large quantities (50 mg per litre of culture) in a highly purified form ( 95%). The purified protein gave a single band at50 kDa on SDS/PAGE.

Two soluble enzymes (FerA and FerB) catalyzing the reduction of a number of iron(III) complexes by NADH, were purified to near homogeneity from the aerobically grown iron-limited culture ofParacoccus denitrificansusing a combination of anion-exchange chromatography (Seph-arose Q), chromatofocusing (Mono P), and gel permeation chromatography (Superose 12). FerA is a monomer with a molecular mass of 19 kDa, whereas FerB exhibited a molecular mass of about 55 kDa and consists of probably two identical subunits. ...

A gene encoding a eugenol oxidase was identified in the genome fromRho-dococcus sp. strain RHA1. The bacterial FAD-containing oxidase shares 45% amino acid sequence identity with vanillyl alcohol oxidase from the fungus Penicillium simplicissimum. Eugenol oxidase could be expressed at high levels in Escherichia coli, which allowed purification of 160 mg of eugenol oxidase from 1 L of culture. Gel permeation experiments and macromolecular MS revealed that the enzyme forms homodimers.
