In this thesis, the rolling contact wear of 52100 bearing steel and Over-aged Magnesia-Partially-Stabilised Zirconia (OA-Mg-PSZ) ceramic are examined using a newly developed rolling contact wear test rig. The new wear test rig simulates the system geometry of an un-lubricated hybrid (ceramic and steel) ball bearing. The new wear test rig is versatile in that it allows low cost samples to be utilised resulting in a larger number of samples that can be tested.

This thesis is dedicated to study how the charging behaviours of plug-in hybrid vehicles affect the local distribution network. This study focuses on two issues: the power loss and charging cost optimization. The multi-objective particle swarm optimization technique is applied to achieve the optimal charging schedule, resulting in acceptable additional power loss ratio and charging cost.

In this research, a bi-objective hybrid vehicle routing problem is presented and solved by considering alternative paths, various lengths and reliabilities. The first objective function minimizes overall system costs, fuel consumption, and greenhouse gas emissions, whereas the second objective function maximizes the reliability of the entire system with alternative paths and several reliabilities.

The decoy approach against nuclear factor kB (NF-kB) is a useful tool to alter NF-kB dependent gene expression using synthetic oligonucleotides (ODNs) carrying NF-kB specific cis-elements. Unfortunately, ODNs are not stable and need to be extensively modified to be used in vivo or ex vivo. We have previously evaluated the possible use of peptide nucleic acids (PNAs) as decoy molecules. The backbone of PNAs is composed of N-(2-aminoethyl)glycine units, rendering these molecules resistant to both nucleases and proteases.

The siglecs (sialic acid-binding Ig-like lectins) are a distinct subset of the Ig superfamily with adhesion-molecule-like structure. We describe here a novel member of the siglec protein family that shares a similar structure including five Ig-like domains, a transmembrane domain, and a cytoplasmic tail containing two ITIM-signaling motifs. Siglec10 was identified through database mining of an asthmatic eosinophil EST library. Using the Stanford G3 radiation hybrid panel we were able to localize the genomic sequence of siglec-10 within the cluster of genes on chromosome 19q13.

We previously reported that GTS1 is involved in regulating ultradian oscillations of the glycolytic pathway induced by cyanide in cell suspensions as well as oscillations of energy metabolism in aerobic continuous cultures. Here, we screened a yeast cDNA library for proteins that bind to Gts1p using the yeast two-hybrid system and cloned multiple TDH cDNAs encoding the glycolytic enzyme glyceraldehyde-3-phosphate dehydrogenase (GAPDH).

The localization of different classes of alcohol dehydro-genases (ADH) in the brain is of great interest because of their role in both ethanol and retinoic acid metabolism. Conflicting data have been reported in the literature. By Northern blot and enzyme activity analyses only class III ADH has been detected in adult brain specimens, while results from riboprobe in situ hybridization indicate class I as well as class IV ADH expression in different regions of the rat brain.

We isolated two novel 14-3-3 binding proteins using 14-3-3fas bait in a yeast two-hybrid screen of a human brain cDNA library. One of these encoded the C-terminus of a neural specific armadillo-repeat protein, d-catenin (neural plakophilin-related arm-repeat protein or neurojungin). d-Catenin from brain lysates was retained on a 14-3-3 affinity column.

Multiple ankyrin repeat motif-containing proteins play an important role in protein–protein interactions. ANKHD1 proteins are known to possess multiple ankyrin repeat domains and a single KH domain with no known function. Prion diseases are characterized by the accumulation of a pathogenic mis-folded form of a prion protein (PrP) encoded by the Prnpgene in humans. In the present study in zebrafish, two transcripts and the corresponding genes encoding prion proteins, PrP1 and PrP2, related to human PrP have been characterized with a relatively divergent deduced amino acid sequence, but a well preserved overall organization of structural prion pro-tein motifs.
