Activated charcoal (AC) is highly adsorbent and is often used to promote seedling growth in plant tissue culture; however, the underlying molecular mechanism remains unclear. In this study, root and leaf tissues of 10-day-old seedlings grown via immature embryo culture in the presence or absence of AC in the culture medium were subjected to global transcriptome analysis by RNA sequencing to provide insights into the effects of AC on seedling growth.

Five different emasculation methods viz. standard technique for wheat x maize crosses, early pollinations on non-emasculated spikes, clipped glume facilitated emasculation, plastic bag emasculation and hot water emasculation were compared. Clipped glume facilitated emasculation resulted in lowest caryopses formation frequency. Early pollination on non-emasculated spikes gave reasonably high embryo formation frequency while omitting tedious emasculation steps.

Fifty seed of sorghum, wheat and pearlmillet from infected seed lot having characteristics symptom of seed borne fungi. Seed of such symptoms got infected of Curvularia lunata, Bipolaris sorghicola and B. sorokiniana infected seed of sorghum wheat and pearlmillet. Histopathology of apparently in infected seeds confined to mycelium fragments of Curvularia lunata in seed coat and endosperm, embryonic region of sorghum, wheat and pearlmillet seed. Sorghum and pearlmillet seed indicates the presence of brown coloured thicks mycelial fragment of Bipolaris sorghicola in pericarp and embryo region.

Wheat is the second most produced staple food crop in world. An efficient method for in vitro micro propagation and genetic transformation of wheat are crucial for both basic and applied research. The present study was initiated with a view to develop a regeneration system in 4 genotypes of wheat cultivars HD2888, HD2329, DBW148, and DBW88 as a prerequisite to transformation. The mature embryos were excised from seeds and cultured on MS medium supplemented with high and low concentrations of cytokinins and auxins respectively.
